山羊B4GALNT2基因的cDNA克隆及组织表达研究 被引量:3

摘要 根据绵羊β-1,4-N-乙酰氨基半乳糖转移酶2(B4GALNT2)(登录号:KC175557)的mRNA序列设计引物,通过RT-PCR技术对高繁金堂黑山羊和单胎藏山羊卵巢组织B4GALNT2基因c DNA进行克隆、序列分析;采用Real-time PCR技术对发情前期山羊卵巢、子宫、输卵管、垂体、肝脏组织中的B4GALNT2基因的表达进行研究。结果表明:山羊B4GALNT2基因编码区全长为1 521 bp,共编码506个氨基酸,2个品种间有6处碱基差异,导致4处氨基酸差异。B4GALNT2基因mRNA在2个山羊品种卵巢、子宫、输卵管、垂体、肝脏中均有表达,在金堂黑山羊子宫和输卵管的表达量显著高于藏山羊(P<0.05)。说明B4GALNT2基因在动物进化中比较保守,与山羊多羔性状的相关性有待进一步研究。
