豫医卷毛大鼠无毛基因cDNA序列的比较生物学分析

摘要 目的:对豫医卷毛大鼠的无毛(Hr)基因进行比较生物学分析。方法:采用PCR方法对豫医卷毛大鼠Hr基因的c DNA序列进行克隆、测序,并与BN大鼠、小家鼠、野猪、家牛、普通猕猴、人、黑腹果蝇和桔小实蝇的Hr基因c DNA序列及其编码产物的氨基酸序列进行同源性比较和系统遗传学分析。结果:获得了豫医卷毛大鼠Hr基因全长5 232 bp的c DNA序列(Gen Bank登录号:KR109217)。豫医卷毛大鼠Hr c DNA与BN大鼠、小家鼠、野猪、家牛、普通猕猴、人、黑腹果蝇和桔小实蝇的同源性分别为99.8%、87.3%、67.3%、70.2%、80.8%、79.8%、29.8%和24.9%,氨基酸序列的同源性分别为100.0%、92.9%、75.4%、74.8%、76.0%、77.8%、7.4%和11.7%。结论:Hr基因在哺乳动物中具有一定的保守性,与昆虫等非哺乳类动物之间具有较大的差异性。 Aim: To study the difference of hairless( Hr) gene and its coding between Yuyi curly hair rats and other species. Methods: PCR technique was developed to clone the sequence of c DNA of Hr gene from Yuyi curly hair rats,and biologic characters of nucleotide and amino acid sequence encoding Hr gene in different species( rattus norvegicus,mus musculus,sus scrofa,bos taurus,macaca mulatta,homo sapiens,D. melanogaster and Bactrocera dorsalis) were analyzed. Results: The full-length of c DNA sequence of Hr gene from Yuyi curly hair rats was 5 232 bp( Accession Number:KR109217). The Hr gene nucleotide homology of Yuyi curly hair rats to rattus norvegicus,mus musculus,sus scrofa,bos taurus,macaca mulatta,homo sapiens,D. melanogaster and Bactrocera dorsalis was 99. 8%,87. 3%,67. 3%,70. 2%,80. 8%,79. 8%,29. 8% and 24. 9%,and that of amino acid sequences was 100. 0%,92. 9%,75. 4%,74. 8%,76. 0%,77. 8%,7. 4% and 11. 7%,respectively. Conclusion: A certain degree of conservation of Hr gene exists among different mammalian taxons,but there is some specificity from non-mammals.
