高效液相色谱法测定连花清瘟颗粒中连翘酯苷A含量

Content Determination of Forsythoside A in Lianhua Qingwen Granules by HPLC

目的 建立连花清瘟颗粒中连翘酯苷A 含量的高效液相色谱测定方法,为更好地控制连花清瘟颗粒质量提供参考.方法 采用十八烷基硅烷键合硅胶色谱柱,流动相为乙腈-0.4%冰醋酸（15∶85）,流速为1 mL/min,柱温为室温,检测波长330 nm.结果 连翘酯苷A 在0.1～1.4 μg 范围内线性关系良好,回归方程为Y=1240X-16.82（r=0.999 9）,平均回收率为101.53%,RSD=1.71%.结论 本方法准确、可靠、重复性好,能测定连花清瘟颗粒中连翘酯苷A 含量,可作为连花清瘟颗粒质量控制的含量检测方法.

Objective To provide a better reference for the quality control of Lianhua Qingwen Granules by establishing the method for the content determination of Forsythoside A in Lianhua Qingwen Granules by HPLC. Methods The chromatographic conditions were Agilent C18 as chromatographic column, acetonitrile-0.4% acetic acid(15∶85) as the mobile phase; the flow rate was 1.0 m L/min; the column temperature was at room temperature; the detection wavelength was 330 nm. Results Forsythoside A was in a good linear range between 0.1–1.4 μg; regression equation was Y=1240X-16.82(r=0.999 9); the average recovery was 101.53%, RSD=1.71%.Conclusion The method is accurate, reliable, and with good repeatability, which can be used for the content determination of Forsythoside A in Lianhua Qingwen Granules and the quality control of Lianhua Qingwen Granules.
