摘要
目的:脾虚动物水液代谢特征研究。方法:饮食不节加劳倦过度法制作大鼠脾虚模型。造模结束后,对模型组大鼠进行表观症状和体内指标评价,记录游泳时间,6 h排尿量及24 h进食量。测定胃残留率、小肠推进率。测定水液代谢相关指标,白蛋白(ALB)、抗利尿激素(ADH)及红细胞膜Na+-K+-ATP酶水平。测定血浆中D-木糖、环磷酸腺苷(c AMP)、环磷酸鸟苷(c GMP)、c AMP/c GMP、甲状腺激素T3、17羟皮质类固醇(17-OHCS)。结果:脾虚模型表现出中医经典的脾气虚动物症状,表现为游泳时间减少、食量减少、体重下降、倦怠懒动、皮毛干枯、粪便时软时溏、眯眼、弓背。模型动物排尿量、血浆ALB含量、红细胞膜Na+-K+-ATP酶水平均低于正常动物,ADH无显著变化。脾虚动物的胃残留率升高、小肠推进率降低和血浆D-木糖含量下降,说明此次复制的脾虚动物模型符合脾气虚证候。模型组和空白组相比,c AMP,c GMP,c AMP/c GMP,T3,17-OHCS含量无统计学差别。结论:所复制的脾虚大鼠模型通过体内指标的测试排除了肾虚的可能性。而对体内水液代谢的指标测定分析表明,脾虚后水液代谢紊乱,与白蛋白(ALB)及红细胞膜Na+-K+-ATP酶活性降低有关,具有水肿的趋势,这与临床症状相一致。
Objective: To study the characteristics of water metabolism of rats with spleen deficiency. Methods: The spleen deficiency model of rats was made by the methods of improper diet and over exercise. After the formation of spleen deficiency,the apparent symptom,the body index,the food intake in 24 h,the swimming time,the urine in 6 h,the gastric emptying and the intestinal propulsive rates of rats were evaluated. ALB,ADH and the level of Na+- K+- ATPase relating to the water metabolism were determined. The contents of the D- xylose,c AMP,c GMP,c AMP / c GMP,T3,17- OHCS in plasma were determined. Results: The model rats showed the classic spleen deficiency symptom of TCM. The model rats appeared that the decreased swimming time,loss of appetite,loss of weight,the dryness of fur,loose stool,squinting and arched the back. The volume of urine,ALB and the level of Na+- K+- ATPase of the model rats were lower than those of the normal rats. ADH showed no significant change. The gastric emptying rate of the model increased. The intestinal propulsive rate and the content of the D- xylose of the model decreased. It illustrated the replication of the animal model according with the spleen deficiency of TCM. Compared with the model group and the control group,the contents of c AMP,c GMP,c AMP / c GMP,T3,17- OHCS showed no statistical difference. Conclusion: The replication of the spleen deficiency model rats excluded the possibility of the kidney deficiency by in vivo test. Water metabolism in vivo indexes showed the disorder of water metabolism caused by the spleen deficiency was relating with the decreased content of ALB and the level of Na+- K+- ATPase. The animal model also had the tendency of edema in accord with the clinical symptom.
