摘要
目的:制备一种稳定可靠的糖尿病足溃疡大鼠模型。方法:将40只180~200 g雄性SD大鼠随机分为4组,第1组、第2组高脂高糖饲料喂养1个月,分别腹腔注射STZ40 mg/kg、腹腔注射STZ50 mg/kg;第3组、第4组普通饲料喂养1周,分别腹腔注射STZ60 mg/kg、腹腔注射STZ70mg/kg。将50只80~100 g雄性SD大鼠随机分为3组,第5组、第6组各20只高脂高糖饲料喂养1个月,分别腹腔注射STZ40 mg/kg、腹腔注射STZ50 mg/kg。第7组10只空白对照组,普通饲料喂养。按时监测大鼠大体情况及死亡率,观察28 d。选取稳定的糖尿病大鼠模型10只和第7组空白对照组大鼠10只在无菌条件下剪去造模区皮肤,深达筋膜。建立糖尿病足溃疡模型,计算两组愈合率。结果:第5组从第3天开始到第28天观察结束,血糖浓度明显高于空白对照组,血糖值均〉16.7 mmol/L。死亡率为25%。与对照组比较,实验组大鼠创面愈合率与空白对照组比较愈合延迟。结论:80~100 g的雄性SD大鼠高脂高糖饲料喂养1个月,腹腔注射STZ40 mg/kg制备糖尿病足溃疡大鼠模型最为稳定可靠。
Objective: To prepare a stable and reliable diabetic foot ulcer model in rats. Methods: 40 male SD rats( 180 ~ 200g) were randomly divided into 4 groups. Group 1 and group 2 were fed high fat and sugar for a month and given abdominal cavity injection STZ respectively 40 mg / kg and 50 mg / kg. Group3 and group 4 were fed normally and given abdominal cavity injection STZ respectively 60 mg / kg and 70 mg / kg. 50 male SD rats( 80 ~ 100 g) were randomly divided into 3 groups. Group 5 and group6 each had 20 rats,feeding high fat and sugar for a month and abdominal cavity injection STZ respectively 40 mg / kg and STZ 50 mg / kg. Blank control group( group 7) 10 rats were fed normally. The general situation on time monitoring rats and mortality were compared,observing for 28 days. 10 rats of the stable diabetic models and 10 of blank control group were selected,under aseptic conditions to build mold areas of skin,deep to fascia. Diabetic foot ulcer model was set up,calculating rate of two groups of healing. Results: From day 3 to 28 d in group 5,blood sugar concentration was significantly higher than that of the blank control group.The blood sugar value 16. 7 mmol / L. Mortality was 25%. Compared with control group,experimental group rats delayed healing compared with blank control group. Conclusion: 80 ~ 100 g male SD rats which are fed high fat and sugar for a month and given intraperitoneal injection STZ 40 mg / kg is the most stable and reliable method for the diabetic foot ulcer model.
