电针内关穴对肝郁胸痹证大鼠IL-1、IL-2、IL-8表达的影响

Influence of Electro-acupuncture at Neiguan( PC6) on Expressions of IL-1,IL-2 and IL-8 of Rats with Chest Bi Sydnrome with Liver Depression

目的:通过对比观察电针内关穴对肝郁胸痹证大鼠白细胞介素-1(IL-1)、白细胞介素-2(IL-2)、白细胞介素-8(IL-8)表达的影响,探讨电针内关穴对肝郁胸痹证的治疗机制。方法:选取雄性健康SPF级大鼠40只,分为4组:空白组(n=10)、模型组(n=10)、电针治疗组(电针内关穴)(n=10)、电针对照组(电针非经非穴)(n=10)。除空白组外,其他组均以高脂饮食加注射垂体后叶素,并配合慢性束缚及游泳建立肝郁胸痹证大鼠模型,内关穴组与非经非穴组采用电针治疗,连续14 d。采用酶联免疫吸附法检测各组大鼠血清IL-1、IL-2、IL-8表达。结果:与空白组比较,模型组的IL-1、IL-2、IL-8含量明显升高(P<0.01);与模型组比较,电针治疗组和电针对照组的IL-1、IL-2、IL-8含量降低(P<0.01)。与电针对照组比较,电针治疗组的IL-1、IL-2、IL-8含量降低(P<0.05)。结论:电针内关穴能对肝郁胸痹证所产生的炎症因子有特异性调控作用,对肝郁胸痹证有一定的治疗作用。

Objective： To observe the effects of electro- acupuncture at Neiguan（ PC6） on expressions of interleukin- 1（ IL-1）,interleukin- 2（ IL- 2） and interleukin- 8（ IL- 8） in rats with chest Bi symptom with liver depression and to explore the therapeutic mechanism. Methods： 40 SPF male rats were randomly divided into 4 groups： a control group（ n = 10）,a model group（ n =10）,a electro- acupuncture treatment group[electroacupuncture at Neiguan（ PC6） ]（ n = 10） and a electro- acupuncture control group（ electroacupuncture at non- acupoint）（ n = 10）. Except the control group,all other groups were given high fat diet and were injected Pituitrin. Furthermore,they were trapped and forced swimming in order to establish the rat model of depression and obstruction of the heart collaterals and vessels. The Neiguan group received electro- acupuncture treatment for 14 days. After that,the IL- 1,IL- 2 and IL- 8 contents of all rats were detected by ELISA. Results： Comparing with control group,the IL- 1,IL- 2 and IL- 8 levels in model group were significantly high（ P〈0. 01）. Comparing with model group,the IL- 1,IL- 2 and IL- 8 levels in the electro- acupuncture treatment group and electro- acupuncture control group were significantly low（ P〈0. 01）. Comparing with electro- acupuncture control group,the IL- 1,IL- 2 and IL- 8 levels in the electroacupuncture treatment group were lower（ P〈0. 05）. Conclusion： The study indicates that electro- acupuncture at Neiguan（ PC6） could specifically regulate inflammatory factor in rats with depression and obstruction of the heart collaterals and vessels.
