摘要
目的探讨芹菜素改善烟草烟雾提取物诱导的血管内皮细胞损伤的机制。方法培养人脐静脉内皮细胞株,给予烟雾提取物(CSE)模拟吸烟环境和不同浓度芹菜素。实验分为正常对照组、CSE组、芹菜素组,检测细胞存活率、凋亡率、细胞内活性氧ROS水平,以及细胞上清液中IL-6、TGF-β、INF-γ各因子的表达。结果芹菜素在24 h和48 h均能显著缓解不同浓度CSE诱导的内皮细胞存活率的降低,降低不同浓度CSE诱导的内皮细胞的凋亡率,同时芹菜素可抑制CSE诱导的细胞内ROS水平,以及上清液中炎性因子IL-6、TGF-β、INF-γ的升高,差异均有统计学意义(P<0.05,P<0.01)。结论芹菜素以血管内皮细胞凋亡和炎性反应为标靶,抑制吸烟引起的血管内皮细胞损伤,为临床治疗提供理论依据。
Objective To investigate the mechanism of apigenin in endothelial cell damage induced by cigarette smoke extract(CSE).Methods Human umbilical vein endothelial cells were cultured with CSE simulating the smoke environment and were given varying concentrations of apigenin.The cells were divided into normal control group,CSE group and apigenin group.Cell viability,apoptosis rate,intracellular ROS level,and IL-6 TGF-β,INF-γ of cell supernatant were detected.Results Apigenin could significantly improve the survival rates of endothelial cells with different concentrations of CSE at 24 h and 48 h,and reduce the rate of apoptosis induced by different concentrations of CSE.Apigenin could reduce the elevated levels of ROS induced by CSE and IL-6,TGF-β,INF-γ,there were significant difference(P〈0.05,P〈0.01).Conclusion Apigenin can inhibit endothelial cell damage caused by smoking through apoptosis and inflammatory reactions,and provide a theoretical basis for clinic.
