摘要
目的探讨脓毒症模型大鼠急性肺损伤(acute lung injury,ALI)时血清降钙素原(procalcitonin,PCT)、C-反应蛋白(C-reactive protein,CPR)和抵抗素水平变化的临床意义。方法wistar雄性大鼠45只随机分为对照组、假手术组和模型组各15只。尾静脉注射脂多糖(LPS)10mg/kg制成脓毒症动物模型,分别于6、12和24h后活杀,测定肺湿/干重比值,免疫荧光层析法检测血清PCT水平,胶体金法检测血清CRP水平和ELISA法检测血清抵抗素水平,并进行对比性分析。结果模型组大鼠血清PCT和抵抗素水平较之对照组和假手术组均明显增高(P均〈0.001),血清CRP较之对照组和假手术组差异无统计学意义(P均〉0.05)。结论脓毒症模型大鼠ALI时血清PCT和抵抗素明显增高,PCT和抵抗素参与脓毒症大鼠的ALI过程,故血清PCT和抵抗素是早期诊断脓毒症的有价值指标。
Objective To explore clinical significance of changing of procalcitonin (PCT), C-reactive protein (CRP) and resistin levels in the model acute lung injury (ALI) rats with sepsis. Methods Forty-five Wistar rats were randomly assigned to three groups, a control group, a sham group and a model group. Sepsis was induced by intravenous injective of LPS (10 mg/kg) and those animals were killed after 6, 12 and 24 hours. The lung wet/ dry weight ratio was detected, the serum PCT level was detected by immunofluorescence assay, the CRP level was detected with Colloidal gold assay, the serum resistin level were measured with ELISA and were done compared analysis. Results Compared with the control group and the sham group, the serum levels of PCT and resistin in model group were significantly higher (P all 〈 0.001). But no significant difference in CRP level was found (P all 〉 0.05). Conclusion PCT and resistin significantly increased in ALl with sepsis rats, PCT and resistin possibly participate the process of sepsis rats induced ALI, so serum PCT and resistin can be as valuable marker to early diagnose sepsis.
