人类SEPT6蛋白对胰腺导管癌细胞系PANC-1迁移和侵袭能力的影响

摘要 Septin家族是在大部分真核生物中都存在的一类蛋白质,研究表明它们除了和细胞中的胞浆移动相关之外还与胞膜运动、胞吞胞吐等过程联系密切.本文旨在通过对该家族中SEPT6的探究,了解它与癌症之间的某些联系.我们通过Western blot和病理组织免疫组化的实验证实了SEPT6在胰腺导管癌中高表达,因此选择胰腺导管癌来源的PANC-1细胞作为研究对象.通过细胞划痕实验,我们发现过量表达SEPT6有助于加速细胞迁移,相反,消减SEPT6会明显遏制划痕的愈合.同样,在Transwell实验中,我们也证实了SEPT6和PANC-1的侵袭能力紧密相关,这些实验证据都提示该蛋白和胰腺导管癌的恶性程度成正相关. A family of Septins exists in most eukaryotes, and plays a pivotal role during cytokinesis in both fungal and animal cells. In addition, it is involved in the membrane dynamics, the process of endocytosis and exocytosis. In this study, we sought to learn one of the family members, SEPT6, and reveal the relation between SEPT6 and cancer. SEPT6 was found highly-expressed in pancreatic ductal adenocarcinoma through Western blot and immunohistochemical staining of pathological tissues, so the PANC-1 cells derived from pancreatic ductal adenocarcinoma was used as our research model. In the wound healing test, we found that over-expression of SEPT6 accelerated the migration of PANC-1 cells, in contrast, knockdown of SEPT6 significantly inhibited the migration. We also proved that SEPT6 was closely related to invasion ability of PANC-1 through transwell assays.
