摘要
目的:研究人大肠癌鸡胚尿囊膜移植模型的建立方法。方法:将孵育7d的种蛋制备成鸡胚绒毛尿囊膜(CAM)模型,随机分为5组,分别在假气室相对无血管区接种数目为2.5×106、5.0×106、1.0×107、2.0×107的HCT 116细胞,对照组接种等体积PBS缓冲液,继续孵育7d,观察并比较各组鸡胚存活及成瘤情况。结果:各组间鸡胚存活情况无显著差异,5.0×106、1.0×107、2.0×107组成瘤率最高,3组间成瘤率无显著差异(P>0.05)。结论:HCT 116细胞可在CAM上形成移植瘤,最佳接种细胞数为5.0×106。
Objective: To establish a human colorectal cancer xenograft model on the chick embryo chorioallantoic membrane( CAM). Methods: The chick embryo CAM model was made using fertilized chicken eggs incubated for 7days. Those eggs were randomly divided into five groups. Four groups received inoculation of 2. 5 × 106,5. 0 ×106,1. 0× 107,and 2. 0 × 107HCT116 cells in the avascular zones of gas chamber,respectively,while the control group received inoculation of an equal volume of PBS buffer. The survival and tumor development in th ose chicken embryos were evaluated after 7 days of incubation. Results: There was no significant difference in the survival between different groups of chicken embryos. The three groups inoculated with 5. 0 × 106,1. 0 × 107,and 2. 0 × 107HCT116 cells had the highest rates of tumor development. There was no significant difference in the rate of tumor development between the three groups( P 0. 05). Conclusion: The HCT116 cell xenograft tumors can develop on the chick embryo CAM. The optimal number of cells for inoculation is 5. 0 × 106.
