茶树根抗痛风活性部位的筛选研究

摘要 目的:比较茶树根总黄酮和总皂苷对高尿酸血症小鼠尿尿酸、血尿酸、血黄嘌呤氧化酶活性的影响和对抗急性痛风性关节炎作用以及体外对黄嘌呤氧化酶活性抑制作用,探讨茶树根防治痛风的活性部位。方法:用尿酸和氧嗪酸钾ip小鼠造成小鼠高尿酸血症模型,分光光度法检测尿酸水平和黄嘌呤氧化酶活力;以尿酸钠结晶植入法观察茶树根总黄酮和总皂苷抗急性痛风性关节作用;以含酶动力学分析软件的分光光度计在波长290nm处测定茶树根总黄酮和总皂苷对黄嘌呤氧化酶活性的抑制作用。结果:茶树根总黄酮在降血尿酸和促尿酸排泄以及抑制黄嘌呤氧化酶活性上起主要作用,茶树根总皂苷则对急性痛风性关节炎起主要抑制作用。结论:茶树根总黄酮和总皂苷均为茶树根抗痛风的药效部位。 Objective:To study the active fractions of tee root about anti -gout by comparative study the effects of total flavonoids and total saponins of tee root on urine uric acid and blood uric acid and serum xanthine oxidase activity in hyperuricemia mice and the effects on acute gouty arthritic mice and the effects on the inhibition on Xanthine oxidase activity of them in vitro. Methods:We injected oxonic acid potassium and uric acid into the abdominal of mice to made the hyperuricemia in mice,and determined serum uric acid and urine uric acid and serum xanthine oxidase activity by spectrometric method. In addition, Monosodium urate crystals were used to establish gouty arthritis model rats, We detected the inhibition of Swelling degree on the right foot pad part of mice. At last , We determined the Inhibition of the total flavonoids and the total saponin of tee root on xanthine oxidase in vitro by spectrophotometer with enzyme kinetics software in the wavelength of 290nm. Results :The results showed that total flavonoids of tee root could not only reduce the serum uric acid and serum xanthine oxidase activity and increase urine uric acid in hyperuricemia mice, but also inhibit xanthine oxidase in vitro. But total saponin of tea root has few effection on the above bioactivity. On the contrary,total saponin of tea root had much stronger anti acute gout arthritis effect than total flavonoids of tee root. Conclusion:Total flavonoids of tee root and total saponins of tee root are the active fractions about anti - gout of tee root.
