摘要
目的:观察一贯煎单用及与环磷酰胺联用对荷H22肝癌小鼠MMP9、NF-κB表达的影响。方法:雄性昆明小鼠40只,随机分为模型组、化疗组、一贯煎组、一贯化疗联合组。腋下注射H22细胞,建立H22荷瘤模型,一贯煎组造模前两周开始用药[剂量为23g mg/(kg·d)],化疗组[剂量为50mg/(kg·d),隔日1次],一贯化疗联合组造模后用药,造模14d后处死小鼠称重,取瘤组织称重计算瘤指数,取脾组织称重计算脾指数,免疫组化检测MMP9、NF-κB蛋白的表达。结果:化疗组、一贯煎组及一贯化疗联合组均具有明显的抑瘤作用,与模型组比较差异有统计学意义(P<0.05或P<0.01);一贯煎组脾指数增大,与化疗组比较差异有统计学意义(P<0.05);一贯化疗联合组能减少MMP9蛋白的表达,与模型组比较差异有统计学意义(P<0.05);化疗组、一贯煎组、一贯化疗联合组均能减少NF-κB蛋白的表达,与模型组比较差异有统计学意义(P<0.01)。结论:一贯煎、环磷酰胺及其二者联用均具有抗肿瘤的作用,且均能减少NF-κB蛋白的表达而抑制肿瘤细胞的生长,但只有在二者联用的情况下才具有降低肝癌组织MMP9蛋白表达的作用,这可能会减少肝癌细胞的转移。
Objective:To observe the effect of Yiguanjian single and combined with cyclophosphamide(CTX) on MMP9,NF - KB expres- sion in I-I22 tumor- bearing mice. Method:40 male Kunming mice were divided into model group, CTX group, Yiguanjian group and the combination group. Animal models of H22 tumor - bearing mice was induced with the axillary injection of H22 cells, Yiguanjian group was started medication with two weeks before [ 23g/( kg · d) ], CTX group[ 50mg/( kg · 2d) ] and the combination group started medication after modeling. After 14d modeling, killing the mice and weighing the tumor tissue and splenic tissue, calculation of the tumor and spleen index, testing the expression of MMP9, NF - KB protein on immunohistochemistry assay. Result:The CTX group, Yiguanjian group and the combination group have obvious antitumor effects, the difference being statistically significant compared to the model group( P 〈0. 05 or P 〈0. 01 ) ;Yiguanjian group Spleen index increased, the difference being statistically significant compared to the model group ( P 〈0. 05 ) ; The combination group have declined significantly on the expression of MMP9, the difference being statis- tically significant compared to the model group( P 〈 0.05 ) ; Yiguanjian group, CTX group and the combination group all have declined significantly on the expression of NF - KB, the difference being statistically significant compared to the model group(P 〈0. 01 ). Conclusion : Yiguanjian, CTX and union all have antitumor effects, all can reduce the expression of NF - KB protein that inhibit the growth of tumor cells, but only in two were combined can reduced the expression of MMP9 protein in hepatoceUular carcinoma, this may reduce the metastasis of tumor.
