摘要
目的:建立一种大麻活性成分△~9-四氢大麻酚﹙THC﹚及其主要代谢物△~9-氘代四氢大麻酚﹙THCC﹚的液相色谱-串联质谱-多反应监测(LC-MS/MS-MRM)分析方法。方法:毛发经去污处理后,甲醇萃取分离、纯化,采用LC-MS/MS法分析,用正负离子切换模式进行THC与THCC的定量分析。结果:该方法对THC与THCC的线性范围为1~20 ng/mg﹙r〉0.997﹚,检测限分别为0.5 ng/mg和0.25 ng/mg,定量限分别为1ng/mg与0.5 ng/mg。日内和日间的精密度分别为1.2%~8.8%和1.9%~9.5%,准确度为86.9%~113.1%。结论:建立的LC-MS/MS分析方法快速、灵敏、准确、可靠,适用于毛发中大麻活性物质及其代谢产物的定量测定。
Objective: A liquid chromatography- tandem mass spectrometry (LC/MS/MS) method in multiple reaction monitoring (MRM) mode was developed and validated for the simultaneous determination of A9 -tetrahydrocannabinol (THC) and 11 - nor -9 - carboxy - A9 - tetrahydrocannabinol (THCC) in human hair. Methods : Hair samples after decontamination, was extracted by meth- ylene, purified. LC/MS/MS analysis with polarity switching was then performed to ctuantitate THC and THCC. Results: The linear range was 1 ~ 20 ng/mg ( r 〉 0. 997 ). The limit of detection were 0.5 ng/mg and 0.25 ng/mg for THC and THCC. The limit of quanti- fication were 1 ng/mg and 0.5 ng/mg, respectively. The relative standard deviations of intra and inter- day precisions were 1. 2% -- 8.8% and 1.9% -- 9.5%, respectively. The inter - day accuracy ranged from 86.9%-- 113.1%. Conclusion: The result showed that the method was rapid, sensitive and accurate, and was applicable in the quantitative determination of THC and THCC in human hair.
