摘要
目的:通过实验方法分析槲寄生断面异常绿色的成因并探讨该绿色与质量的相关性。方法:用溶剂提取该绿色成分,采用紫外灯下观察荧光,紫外-可见光吸收波长扫描、薄层色谱等方法判断性质,并用高效液相色谱法测有效成分紫丁香苷的含量。结果:该绿色成分可被95%或无水乙醇冷提,提取液在365 nm紫外光下显红色荧光,在波长665 nm有最大吸收峰。该提取液薄层下相应斑点Rf值及颜色与叶绿素相符合,紫丁香苷含量测定达到药典标准。结论:槲寄生的断面绿色是由于叶绿素大量保留形成的。饮片的紫丁香苷含量不因绿色多少而减少。叶绿素被完好保存并颜色特异可能与加工方法、栽培条件、采收时间和贮藏条件等因素相关,具体原因有待进一步探讨,但药典的有关描述是否需要修订值得商榷。
Objective: To investigate the reasons why the cross section of colored mistletoe herb can be green. Methods: The green components were extracted by solvent,and the property of the components were determined by ultraviolet light,fluorescent light-visible lights' wavelength scanning and thin layer chromatography,and the content of active components was determined by HPLC. Results: The 95% ethanol extract of green colored mistletoe herb pieces contained the green components. The ethanol extract had red fluorescence under 365 nm UV light and the wavelength of the maximum absorption peak was 665 nm. The Rf value and color of spot on extract correspond with chlorophyll. The syringin in the green decoction pieces of colored mistletoe herb reached Chinese Pharmacopoeia standards. Conclusion: The green color in the cross-section of colored mistletoe herb was caused by chlorophyll,and there was no declination of syringin. There should be more studies on the reasons why the chlorophyll was saved so well with the special green color. Whether the related description on China's pharmacopoeia should be revised should be further discussed.
