褪黑素对创伤性失血性休克大鼠脏器损伤保护作用 被引量:1

摘要 目的观察外源性褪黑素(MT)对创伤性失血性休克大鼠肝、肺损伤的保护作用。方法 30只Wistar大鼠随机平均分为三组(n=10),分别为空白对照组(C)、创伤性失血性休克+磷酸盐缓冲液(PBS)复苏组(S)、创伤性失血性休克+褪黑素(MT)复苏组(L)。C组不建立休克模型,S组在休克复苏前静脉注射PBS 1 ml,L组在休克复苏前静脉注射褪黑素(MT)1 mg,休克复苏后观察240 min。酶联免疫法(ELISA)检测血清肿瘤坏死因子-α(TNF-α)及肺组织髓过氧化物酶(MPO)含量的变化,生化法检测血清ALT、AST含量变化。苏木素-伊红(HE)染色法检测肺组织病理学变化。结果两组创伤性失血性休克大鼠肺组织MPO含量均高于对照组(P<0.05),L组大鼠肺组织MPO含量明显低于S组(P<0.05)。两组创伤性失血性休克大鼠血清TNF-α含量均高于对照组(P<0.05),L组大鼠血清TNF-α含量明显低于S组(P<0.05)。两组创伤性失血性休克大鼠血清ALT及AST含量均高于对照组(P<0.05),L组大鼠血清ALT及AST含量明显低于S组(P<0.05)。结论 MT能够减轻创伤性失血性休克所导致的大鼠肝、肺损伤,降低炎症反应,利于休克的复苏及脏器的保护。 Objective To investigate the protective effects of exogenous melatonin( MT) on organ injury after hemorrhagic shock in rats. Methods Thirty adult male Wistar rats were randomly assigned into three groups: control group( Group C),hemorrhagic shock and resuscitation with PBS group( Group S),hemorrhagic shock and resuscitation with MT group( Group L). Group C underwent the surgical procedure but were not bled. Group S were injected PBS( 1 ml before resuscitation),and Group L were injected MT( 1 mg before resuscitation). Tumor necrosis factor-α( TNF-α) in serum and myeloperoxidase( MPO) in lung tissue were assayed with ELISA. The serum contents of ALT and AST were detected with biochemical methods. The lung pathological change was examined by hematoxylin and eosin( HE) staining. Results The lung contents of the MPO in Group S and Group L were significantly higher than that in Group C( P〈0. 05),and the MPO in Group S was significantly lower than Group L( P〈0. 05). The serum contents of TNF-α in Group S and Group L were significantly higher than that in Group C( P〈0. 05),and the TNF-α in Group S was lower than that in Group L( P〈0. 05). The serum contents of ALT and AST in Group S and Group L were significantly higher than that in Group C( P〈0. 05) and that of ALT and AST in Group S were significantly lower than that in Group L( P〈0. 05). Conclusion Melatonin may reduce lung and hepatic injury in rats after hemorrhagic shock,and therefore reduce the inflammatory response.
