摘要
目的利用Caco-2和MDCK细胞单层模型探讨石杉碱甲与银杏内酯B的跨细胞转运过程及其机制。方法建立Caco-2和MDCK细胞单层模型,将Caco-2和MDCK细胞分别按密度1×105、5×104个细胞/cm2接种到transwell culture plate inserts上培养,待细胞单层达到一定密度程度后进行转运实验。用高效液相色谱法测定石杉碱甲和银杏内酯B在不同侧面、不同浓度条件的跨单层细胞转运的情况,同时计算累计的透过量。结果在两种不同单层细胞模型上,石杉碱甲与银杏内酯B都有不同程度的吸收和分泌。结论石杉碱甲和银杏内酯B跨细胞转运同时存在吸收和分泌的动力学过程。
Objective To study the transcellular transport of huperzine A and Ginkgolide B. Methods After Caco-2 and MDCK cell monolayer was set up,Caco-2 and MDCK cells( at a density of 1 × 10^5 and 5 × 10^4 cells / cm2) were inoculated onto Transwell Culture Plate Inserts for culture. Transcellular transport tests of huperzine A and ginkgolide B across monolayer were performed. Results There were some absorption and secretion of huperzine A and ginkgolide B across the two kinds of monolayers. Conclusion There are simultaneous dynamic actions of absorption and secretion during transcellular transport of huperzine A and ginkgolide B.
