摘要
目的:探讨老年冠心病患者细胞色素P450 2C19(CYP2C19)基因多态性及代谢表型的分布。方法:应用DNA微阵列芯片法对604例来我院就诊的老年冠心病患者进行CYP2C19基因分型。结果:本研究中检测到CYP2C19的6个不同基因型为:CYP2C19*1/*1,*1/*2,*1/*3,*2/*2,*2/*3,*3/*3,所占比例分别为40.2%、40.9%、6.6%、10.1%、2.0%及0.2%。CYP2C19三种代谢表型:快代谢型、中间代谢型、慢代谢型,所占比例分别为40.2%、47.5%、12.3%。男性:快代谢型占38.0%(168/442),中间代谢型占48.9%,(216/442),慢代谢型13.1%(58/442);女性:快代谢型占46.3%(75/162),中间代谢型占43.8%,(71/162),慢代谢型9.9%(16/162)。结论:604例老年冠心病患者中CYP2C19的基因型分布及代谢表型,与文献报道的中国汉族健康人群基本一致,其在男女间分布差异无统计学意义。
Objective: To investigate the genetic polymorphisms of and metabolic phenotype cytochrome P450 2C19( CYP2C19) in senile patients with coronary heart disease. Methods: The genotypes of CYP2C19 in604 senile patients with coronary heart disease were determined by using DNA microarray method. Results: Six genotypes of CYP2C19 were detected in this study with CYP2C19*1 /*1 accounting for 40. 2%,CYP2C19*1 /*2 40. 9% 、 CYP2C19*1 /*3 6. 6%, CYP2C19*2 /*2 10. 1%, CYP2C19*2 /*3 2. 0%, and CYP2C19*3 /*3 0. 2%. There were three types of metabolic phenotype including rapid metabolism type,intermediate type,and slow type accounting for 40. 2%,47. 5%,and 12. 3%,respectively. In male patients,the percentage of the three types of metabolic phenotype was 38. 0%( 168 /442),48. 9%( 216 /442),13. 1%( 58 /442) respectively,and 46. 3%,43. 8%( 71 /162),9. 9%( 16 /162) respectively in female patients. Conclusion: The distribution of CYP2C19 genotypes in 604 senile patients with coronary heart disease is consistent with that in Han population of different age. There is no significant difference between male and female patients.
