摘要
为了增强蛋白类药物的活性和寿命,应用物理交联技术制备了壳聚糖(CS)/京尼平(GP)/明胶(G)/β-甘油磷酸钠(β-GP)共混的温敏性水凝胶,采用红外光谱(FT-IR)、透射电镜(SEM)及降解实验对壳聚糖水凝胶的理化性能进行了表征;并研究了其对牛血清蛋白(BSA)的释放规律.结果显示:壳聚糖温敏性水凝胶的凝胶时间较短,约为50-126s;透射电镜显示4种物质相互交联为互通的多孔网络,结构致密均一;壳聚糖水凝胶具有一定的降解能力;BSA释放时间长达170h,最终释放率为76%-88%,蛋白释放曲线符合药物释放模型.
In order to enhance the activity and life-span of protein,which was applied a thermosensitive chitosan(CS)/genipin(GP)/genlatin(G)/β-glycerophosphate(β-GP)hydrogel was developed using physical crosslinking method.The physicochemical properties of chitosan hydrogel was confirmed by infrared spectroscopy(FT-IR),transmission electron microscopy(SEM)and degradation test,its ability to release BSA.The results showed that the gel time of chitosan thermosensitive hydrogel is short,about 40—140s;The SEM revealed that the four substances crosslinked to porous network with interworking and dense homogeneous;Chitosan hydrogel has a certain ability to degrade;The BSA release time up to 170 hand the final release rate was 76%—88%,Protein release profiles are consistent with the model.
