摘要
目的:观察化瘀通络灸法对皮质下血管性痴呆(SVD)模型大鼠额叶皮质神经元及皮质下白质髓鞘超微结构的影响。方法:4-VO法复制SVD大鼠模型,配合跳台实验确认模型成功,分为正常组、假模型组、模型组、药物组、艾灸组,造模1个月后每组分别治疗,一疗程后取材,电镜下观察各组大鼠额叶皮质及皮质下白质超微结构的变化。结果:与正常组和假模型组相比,模型组皮质神经元明显变性坏死,数量减少;与模型组相比,药物组和艾灸组皮质神经元形态较完整,核膜光滑,胞质内细胞器较为正常。与正常组和假模型组相比,模型组皮质下白质髓鞘出现明显的致密层分离,鞘壁疏松,甚至形成空泡;与模型组相比,药物组和艾灸组皮质下白质髓鞘染色较深,鞘壁致密。结论:化瘀通络灸法可修复SVD大鼠损伤的额叶皮质神经元以及皮质下白质髓鞘,为灸法治疗SVD提供了理论依据。
Objective: To study the effect of Huayu Tongluo moxibustion on the ultrastructure of frontal cortex neurons and subcortical white matter myelin sheath of SVD rats. Methods: SVD rat models were established in accordance with 4- VO method and confirmed by the jumping experiment. Then the rats were divided into a normal group,a model group,a sham model group,a medicine group and a moxibustion group. The treatment started one month later. After one course of treatment,Observe the changes of ultrastructure of the cortex and subcortical white matter of the rats' frontal lobes in each group with the electron microscope. Results: Compared with the normal group and the sham model group,it was discovered that the cortex neurons in model group were degenerative and necrotic,and the number was decreased. Compared with the model group,the neurons in medicine group and moxibustion group were morphologically intact with smooth membrane and the cell organs in cytoplasm were normal. Compared with the normal group and the sham model group,it was discovered that the myelin sheath layer of model group began to separate each other leading to looser sheath cuticle and some vacuoles while the myelin sheath color in medicine group and moxibustion group was stained deeper with more compact sheath cuticle compared with the model group. Conclusion: Huayu Tongluo moxibustion provides theoretical basis for treating SVD by moxibustion therapy.
