摘要 目的建立葎草药材的HPLC指纹图谱,为评价葎草药材的质量提供科学而可靠的依据。方法采用高效液相(HPLC)指纹图谱技术分析葎草药材,经实验确定色谱条件为:采用Agilent HC C_(18)色谱柱(4.6 mm x 250 mm,5μm),以甲醇-0.2%磷酸水溶液作为流动相系统进行梯度洗脱,流速为1.0 m L/min,柱温为35℃,检测为波长340 nm,进样量为10μL。利用中药色谱指纹图谱相似度评价系统对测得的10个批次葎草药材HPLC谱图进行比较分析。结果建立了葎草的HPLC指纹图谱,10个批次的葎草药材相似度都在0.9以上,以大波斯菊苷为参照峰,确立了葎草指纹图谱中的15个共有峰。结论所建立的葎草指纹图谱测定方法简便、稳定性和重复性均良好,可用于葎草药材的常规质量控制检查。 OBJECTIVE To establish an analytic method of the chromatographic fingerprint on Humulus scandens by RP-HPLC for its quality control. METHODS The HPLC chromatographic fingerprints of Humulus scandens were obtained with Agilent HC C18column( 4.6 mm×250 mm,5 μm) by gradient elution solvent system composed of methanol and 0.2% phosphoric acid. The flow rate was 1.0 m L/min,the column temperature was maintained at 35 ℃,and the detector wavelength was set at 340 nm.Using the software of Similarity Evaluation System for Chromatographic Fingerprint of TCM( Version 2004A) to analyze and compare the chromatograms of 10 different batches of Humulus scandens.RESULTS The correlated coefficients of similarity higher than 0.90 were found in fingerprints between different samples of Humulus scandens. Fifteen common peaks were selected in the fingerprint peaks. CONCLUSION The method is convenient,stable and containing good reproducibility,which may be used in regular quality control check of Humulus scandens.
