Simultaneous determination of two effective components in Paidu capsule by HPLC

目的采用HPLC法测定排毒胶囊中大黄素和大黄素甲醚的含量。方法采用Wondisol C18色谱柱（250 mm×4.6mm,5μm）,柱温35℃,流动相为甲醇-0.1%磷酸溶液,以1.0 mL·min^-1流速梯度洗脱,检测波长254 nm。结果大黄素和大黄素甲醚的线性范围分别为7.0-105.0、2.0-30.0μg·mL^-1（r分别为0.9999、0.9992）;加样回收率分别为102.2%、99.2%（n=6）。结论所用方法操作简单、结果准确、重复性好,可有效地控制该制剂的内在质量。

OBJECTIVE To establish an HPLC method for simultaneous determination of emodin and physcion in Paidu capsule by gradient elution. METHODS The quantitative analysis was carried out on a column of Wondisol C18（ 250 mm × 4. 6 mm,5 μm） by a gradient elution using methanol and 0. 1% phosphoric acid as mobile phase at a flow rate of 1. 0 m L·min^- 1at 35 ℃. The eluate was detected by multiple wavelength of 254 nm. RESULTS According to the regression equation,the linear ranges of emodin and physcion fell within the ranges of 7. 0- 105. 0 μg·mL^-1（ r = 0. 9999） and 2. 0- 30. 0 μg·mL^-1（ r = 0. 9992）. The average recoveries were102. 2%（ n = 6） for emodin and 99. 2%（ n = 6） for physcion. CONCLUSION This method is simple,accurate and reliable,and provide a reference standard for the quality control of Paidu capsule.
