半夏SSR分子标记的开发及四种叶型半夏的初步鉴定 被引量:4

摘要 采用磁珠富集法,以芍药叶型半夏作为开发基因组SSR分子标记的材料,利用获得的部分SSR引物进行了4种叶型半夏品种的初步鉴定工作,共筛选到841条半夏基因组序列,其中可用来开发SSR引物的序列有406条,最终成功设计出231对半夏SSR引物,随机挑选62对SSR引物。通过多态性分析,筛选到7对可进行四种表型鉴定的SSR分子标记。结果极大地丰富了半夏基因组信息,获得的SSR序列的重复次数平均值大于20,具有较好的多态性信息显示潜力,为下一步构建半夏指纹图谱和遗传多样性分析等研究奠定了基础。 A magnetic beads enrichment strategy was used to develop the SSR molecular markers in Pinellia ternata ,in order to distinguish the four different leaf type of Pinellia ternata preliminarily. The research got 841 sequences of Pinellia ternata and 406 of them could be used to design the SSR primers and successfully designed 206 pair of SSR primers. Then 62 SSR primers were picked to distinguish four different leaf type of Pinellia ternata and 7 of them could distinguish the four varieties of Pinellia ternata. The SSR sequences had great potential in revealing of polymorphism information and our research greatly enriched nucleotide information of Pinellia ternata, which provided a data and emperical basis for a further study of PineUia ternata.
