摘要
目的研究TH9在大鼠肺纤维化中的表达及其与TGF-β、HGF的关系。方法 SD大鼠随机分三组:对照组(N)、博莱霉素组(B)和地塞米松组(D),每组各30只。造模完成后分别于第7、14、28天随机各处死10只,镜下观察各组肺组织形态学变化、检测肺组织羟脯氨酸(HYP)含量,荧光PCR法检测肺泡灌洗液PU.1、HGF水平,ELISA法检测外周血TGF-β水平。结果 1、HE染色示随时间进展B、D组肺组织由肺泡炎性变发展为纤维性变;HYP的含量变化随时间升高。2、B、D组PU.1、TGF-β水平随时间进展均升高。3、B组HGF水平随纤维化进程降低,D组则随时间变化呈降低复升高的趋势。结论 Th9细胞通过其特异性转录因子PU.1及与HGF、TGF-β的相互作用参与了大鼠肺纤维化的过程。
Objective To explore the effect of Th9 cell in rat model of pulmonary fibrosis and the relation- ship with HGF and TGF-β in the process of PF. Methods 90 SD rats were randomly divided into 3 groups : the control group ( N group, n = 30), the PF model group ( B group, n = 30) and the dexmethasone treatment group ( D group). Rats were sacrificed at the 7th, 14th, 28th days after modeling in batch. Lung pulmonary tissues and the level of hydroxyproline (HYP) were restored to analyze the pathological changes with HE staining. The levels of PU. 1 and HGF in the BALF of rats with pulmonary tuberculosis were detected by real time PCR, and the level of TGF-β in blood was evaluated by ELISA. Results 1. Biopsy of pulmonary tissues showed a dynamic process of change from alveolitis to fibrosis in the B and D groups. The level of HYP in pulmonary tissues got higher with the time prolon- ging. 2. The levels of PU. 1 and TGF-β showed a higher expression in the B and D groups. 3. In the B and D groups, the level of HGF showed a lower expression. Conclusion Th9 has a relationship with PF through HGF, TGFβ and its specific transcription factor PU. 1.
