裙带菜孢子叶岩藻黄质提取工艺与抗氧化活性的研究 被引量:10

Extraction processing and antioxidant activity of fucoxanthin from Undaria Pinnatifida sporophyll

摘要 以裙带菜孢子叶为原料,采用超声波法和酶解法复合提取工艺制备岩藻黄质。考察提取溶剂、料液比、超声时间、酶解时间和酶解温度等因素对岩藻黄质提取量的影响,确定最优条件为:提取剂为乙醇:丙酮(3:1)、料液比1:30(w:v)、超声时间10 min、酶解时间35 min、酶解温度50℃时,岩藻黄质提取量达到最佳(0.75±0.03)mg/g。同时采用电子自旋共振波谱技术检测岩藻黄质对DPPH、羟基自由基的清除率,结果表明,岩藻黄质对DPPH和羟基自由基的清除能力随浓度的升高而增强,对DPPH清除率的IC50为3.93μg/m L,对羟基自由基清除率的IC50为30.07μg/m L,表明岩藻黄质具有较强的抗氧化性。 In the present study, Undaria Pinnatifida(U. Pinnatifida) sporophyll as raw material, complex processes including ultrasonic method and enzymatic hydrolysis was used to extract fucoxanthin. Effects of extraction solvent, ratio of solid and liquid, ultrasonic time, enzymatic hydrolysis time and temperature on extraction amount was analyzed. The results indicated that the best extraction amount(0.75±0.03) mg/g could be reached under the following conditions: ratio of ethanol and acetone was 3:1, ratio of solid and liquid was 1:30(w:v), ultrasonic time was 10 min, enzyme hydrolysis time was 35 min, temperature was 50 ℃. While the antioxidant activity of fucoxanthin was detected by using electron spin resonance(ESR) spintrapping. The result indicated that the DPPH radical and hydroxyl radical scavenging ability enhanced with the increasing concentration of fucoxanthin, and IC50 of DPPH radical scavenging was 3.93 μg/m L, IC50 of hydroxyl radical scavenging was 30.07 μg/m L. These results indicated that fucoxanthin possessed strong antioxidant activity.
