摘要 以菠萝百合花序上胚珠为外植体,接种于3种诱导培养基中,发现加有少量激素的MS+NAA 0.01mg/L及MS+6-BA 0.01mg/L培养基上的胚珠萌芽率明显高于未加激素的MS培养基。以启动培养获得的叶片及小鳞茎纵切块为外植体,接种到3种继代培养基中,发现两种外植体在3种继代培养基上均能分化出不定芽,MS+6-BA 1mg/L+NAA 1mg/L(S3)为最适初次继代培养基。以S3培养基为基础,设计了6种配方进行第二次继代培养研究,发现MS+6-BA 0.5 mg/L+NAA 1 mg/L(T4培养基)上发育的不定芽数最多、质量最好,可作为最适继代培养基。离体生根研究发现:培养基中添加NAA比IBA生根质量更好,最适生根培养基为MS+NAA 0.1mg/L。至此,建立了菠萝百合组培快繁技术体系。 In this study,in order to investigate the tissue culture system of Eucomis pallidiflora,the the ovules were used as explants,and were inoculated on three kinds of induction medium.The results showed that the germination rates of ovules on the MS medium containing 0.01mg/L NAA and MS medium containing 0.01mg/L 6-BA were significantly higher than that on MS medium.The leaves and dices of bulb obtained throught the initiation culture were used as explants to inoculated on three kinds of subculture medium.The adventitious buds were found to be induced from both of these two explants,and MS medium containing 1mg/L 6-BA,and 1mg/L NAA,named S3 medium,was optimum for the first subculture.Then six kinds of mediums were designed for the second subculture based on S3 medium,and the most adventitious buds wigh high quality were found on MS medium containing 0.5mg/L 6-BA,and 1mg/L NAA,named T4 medium,which was optimum for subculture.In the experiment on rooting culture,it was found that the rooting quality through additon of NAA was better than IBA,and MS medium containing 0.1mg/L NAA was optimum for rooting culture.Then the tissue culture and rapid propagation system of E.pallidiflora was established.
