小豆胚轴及胚尖再生和转化体系优化 被引量:2

Optimization for regeneration and transformation system of hypocoty ledonary axis and embryonic tips in adzuki bean(Vigna angularis)

摘要 选用缺铁敏感小豆突变体FMY009的无菌苗下胚轴和胚尖为外植体进行再生和转化,研究了小豆最适胚轴和胚尖再生体系的不定芽伸长培养基和抗生素最适宜的延迟筛选时间。结果表明:小豆胚轴再生不定芽伸长的最佳培养基是1/2 MS B5+1mg/L ZT+0.9mg/L GA3,胚尖再生不定芽伸长的最佳培养基是1/2 MS B5+0.5mg/L GA3,小豆胚轴和胚尖转化体系的抗生素最适延迟筛选时间为7d。 Explants of hypocotyledonary axis and embryonic tips from a sensitive to iron-deficiency mutant FMY009 were used to regenetate and transform.The culture medium of elongating adventitious bud and antibiotics selected time were optimized.The results showed that the optimizer induced media were 1/2 MS B5+1mg/L ZT +0.9mg/L GA3 and 1/2 MS B5+0.5mg/L GA3,respectively.The optimal time of antibiotic selected was 7days.
