短梗胡枝子优良单株组培快繁技术研究 被引量:2

摘要 为实现短梗胡枝子良种的产业化生产,以优良短梗胡枝子单株为材料,研究其组培快繁技术。短梗胡枝子启动培养时,以茎段为外植体,用有效氯1%的次氯酸钠表面灭菌12~14min,成活率高,分化效果好。叶片、叶柄不适合作为组培外植体。茎段增殖的最佳培养基为MS+6-BA 1.0mg/L+NAA 0.2mg/L,最佳生根培养基为1/2 MS+NAA 0.2mg/L+IBA 0.4mg/L,生根率达到了86.3%。试管苗出瓶时,在草炭∶蛭石(2∶1)的栽培基质上成活率最高。 In order to realize the industrialization of improved variety of Lespedeza cyrtobotrya,the tissue culture and rapid propagation technique for excellent individuals were explored.The results showed that using stem segments as explants which were sterilized by NaClO resolution containing 1%available chlorine for 12~14min,the differentiation degree of healing tissue and survival rate of seedlings were high.The leaves and leaf stalks of L.cyrtobotrya were not suitable as explants.The MS medium,containing 1.0mg/L 6-BA,and 0.2mg/L NAA was suitable for proliferation of stem-segment explants.The 1/2MS medium containing 0.2mg/L NAA,and 0.4mg/L IBA was suitable for rooting,and the rooting rate arrived at 86.3%.The seedlings on the cultivation medium composed of moss and vermiculite at the ratio of 2∶1presented the highest survival rate,when the tissue culture seedling were transplanted.
