摘要
为了促进华北紫丁香良种的推广和利用,试验以4个种源的华北紫丁香为材料,研究不同材料在组织培养过程中的分化效果。结果表明:在启动培养时,用有效氯1%的次氯酸钠消毒15~20min,效果最好。华北紫丁香叶片、花蕾、花序轴不适合作为组培外植体,茎段、叶柄、果序轴能分化出愈伤组织,但愈伤组织接种到多种培养基上没能分化出不定芽。以茎段作为外植体时,接种在MS+6-BA 1.0mg/L+NAA 0.2mg/L上芽分化效果最好,芽诱导率达到了86.7%,平均诱导芽数为1.88。分化的芽接种到1/2MS+IBA 0.4 mg/L+NAA 0.2mg/L培养基上,生根效果最好,生根率达到50%。4个种源在芽分化、生根上存在差异,但北京与天津种源之间差异不显著,北京(或天津)种源与山西种源、内蒙古种源之间存在显著性差异。在丁香茎段芽分化中,基本培养基采用MS效果最好,改良MS与WPM效果接近。
In order to promote the popularization and utilization of improved varieties of Syringa oblate Lindl.,the tissue culture method was studied using four provenances of S.oblate Lindl..The results showed that at the stage of initiation culture,treatment with NaClO resolution containing 1% available chlorine for 12-14 min resulted into the optimal sterilization efficacy of the explants.The leaves,flower buds,and inflorescence axises were suitable as explants.Though the callus could be induced from the explants of stem segments,leaf stalks,and inflorescence axises,when inoculated on our mediums,the adventitious buds could not be induced.The MS medium containing 1.0mg.L-1 6-BA and 0.2mg.L-1 NAA was optimal for bud differentiation of the stem segment explants,and the bud induction rate reached 88.6% with 1.88 buds averagely.The optimal medium for rooting of adventitious buds was 1/2MS medium containing 0.4mg.L-1 IBA and 0.2mg.L-1 NAA,and the rooting rate arrived at50%.There was difference in bud differentiation and rooting among these four provenances of S.oblate,but no significant difference was found between Beijing and Tianjin provenances,while there was significant difference between Beijing(or Tianji)provenance and Shanxi,Neimeng provenances.MS medium was optimal for the bud differentiation from the stem segments of S.oblate Lindl.,and the improved MS and WPM medium showed the similar efficacy.
