摘要
以杂交石竹单株自交后代中出现的雄性不育株为材料,茎段为外植体,研究不定芽诱导、不定芽增殖及不定根生成最佳培养条件。结果表明:最佳不定芽诱导培养基为MS+6-BA 1.5mg/L+NAA 0.5mg/L,8d后可诱导出不定芽;最适于不定芽增殖的培养基为MS+6-BA 0.6 mg/L+NAA 0.1 mg/L,增殖倍数可达到10.2倍;最佳生根培养基为MS+NAA 0.3mg/L。将根长6~8cm试管苗移移植到温室炼苗、移栽,成活率达95%以上。
In this study,the rapid propagation technique for a male sterile plant of Dianthus hybridus obtained through selfing was explored.The stem segments were used as explants,then the optimal culuture conditions for adventitious bud induction,adventitious bud multiplication,and root induction were investigated.The results showed that MS medium containing 1.5mg/L 6-BA and 0.5mg/L NAA,was optimal for adventitious bud induction.The optimal medium for adventitious bud multiplication was MS medium containing 0.6mg/L 6-BA and 0.1mg/L NAA.The MS medium containing 0.3mg/L NAA was suitable for root induction.The tissue-cultured plantlets with 6-8cm roots were transferred into greenhouse for seedling adaption,whose survival rate arrived at 95%.This study will provide a theoretical and technical support for developing male sterile lines of D.hybridus.
