逆灸对抑郁模型大鼠血清细胞因子的影响

Effect of Preventive Moxibustion on Serum Cytokines in Depressive Rats

目的:观察逆灸对抑郁模型大鼠血清细胞因子的影响,明确逆灸对抑郁症的预防作用,并进一步探讨其可能机制。方法:40只SD大鼠随机分成正常组、模型组、逆灸组、艾灸组、氟西汀组,每组8只。用慢性不可预见性刺激结合孤养的方法造模;逆灸组在适应性喂养1周后开始艾灸,1次/日,持续3周后进行造模;艾灸组及氟西汀组造模成功后再行治疗3周。采用放射免疫分析法测定大鼠血清IL-1β、IL-6、IL-10含量。结果:抑郁模型组大鼠血清中IL-1β、IL-6含量较正常组显著升高,血清IL-10含量较正常组显著下降;逆灸组、艾灸组及氟西汀组血清中IL-1β、IL-6含量显著下降,逆灸组血清IL-6含量较艾灸组、氟西汀组有显著性差异;逆灸组、氟西汀组血清IL-10含量显著升高,艾灸组血清IL-10含量较模型组无显著性差异,秩检验显示:逆灸组>氟西汀组>艾灸组。结论:艾灸对抑郁症的预防作用优于治疗效果,逆灸通过调节炎性细胞因子和抗炎性细胞因子发挥对抑郁症的预防作用。

Objective： To observe the effect of preventive moxibustion（ PM） on serum cytokines in depressive rats.Methods： Forty male SD rats were evenly randomized into control,model,PM,moxibustion and fluoxetine groups,with 8 rats in each group.Depressive models were established by using chronic unpredictable mild stress（ CUMS） stimulation.PM was given moxibustion for three weeks before model-establishment,and the treatment was conducted once a day,each time for 20 min.Moxibustion and fluoxetine groups were treated for three weeks after successful model-establishment.The contents of interleukin-1β（ IL-1β）,IL-6 and IL-10 in serum were detected by radio-immunoassay.Results： Compared with the control group,the contents of IL-1β and IL-6 in serum in model group were significantly increased,and there were significant differences in serum IL-6 content between the PM and moxibustion and fluoxetine groups（ P〈0.05）;the content of IL-10 in serum in the model group was significantly decreased;compared with model group,preventive moxibustion could significantly decrease the contents of IL-1β and IL-6 in serum,and increase IL-10 level in serum.Rank test showed PM 〉fluoxetine group 〉moxibustion group.Conclusion： The preventive effect of moxibustion on depression is better than that of treatment.Preventive moxibustion can regulate inflammatory cytokines and anti-inflammatory cytokines,which may contribute to its effect on the prevention of depression.
