摘要
目的采用HPLC-ELSD法测定三种不同品种浙贝母:宽叶浙贝、多籽浙贝、狭叶浙贝中贝母素甲和贝母素乙的含量,为浙贝母育种提供实验依据。方法色谱柱Agilent ZORBAX SB-C18(4.6×250 mm,5 um);乙腈-水-二乙胺(70:30:0.03),流速0.8 m L/min;Alltech2000-ELSD:漂移管温度85℃,载气流速为2.2 L/min。结果贝母素甲和贝母素乙线性方程分别为:甲素lg A=0.64 lgm+2.187(r=0.9999),乙素lg A=0.903 lgm+2.082(R=0.9995),贝母素甲在0.925μg^4.625μg,贝母素乙在0.94μg^4.7μg内呈良好的线性关系。贝母素甲和贝母素乙总量平均回收率为98.59%,RSD=1.27%。结论三种浙贝品种中贝母生物碱含量差异显著,除狭叶种浙贝含量未达药典标准,宽叶种和多籽种含量均超过了标准规定,且宽叶种浙贝母含量最佳。结合实验结果及各品种生物学特性建议采用宽叶浙贝育种。
Objeetlve: To determine the contents of peimine and peiminine in three different varieties of Zhejiang Beimu, namely Zhebei of broad leaf, Zhebei of multi-seed and Zhebei of narrow leaf, by HPLC-ELSD, to provide the experimental basis for the breeding of Zbejiang Beimu. Methods: The determined conditions include following aspects: chromatographic colunm : ZORBAX SB-C18 column (4.6 ×250 mm, 5 um ) ; aeetonitrile: water: diethylamine = 70 : 30 : 0.03 ; flow rate: 0.8 mL/min; Alltech2000-ELSD : 85 degrees Celsius of drift tube temperature and 2.2 L/min of cartier gas flow rate. Results: The linear equations of peimine and peiminine were respectively lgA = 0.64 lgrn+2. 187 (r = 0.9999) and lgA=0. 903 lgrn+2. 082(R=0. 9995), the permine was in0. 925 μg -4. 625 p.g and the peiminine in 0.94 Ixg ~ 4.7 μg with a good linear relationship. The total average recovery rate of peimine and peiminine was 98.59 % with RSD= 1.27 %. Conclusion:The contents of alkaloids in these three kinds of Beimu was significantly different. The content of Zhebei of narrow leaf is not up to the standard recorded in Pharmacopoeia, while the contents of Zhebei of broad leaf and Zhebei of multi-seed both exceed the standard and the broad leaf is with best content. Therefore, Zhebei of broad leaf is suggested to breed for the right variety according to the experimental results and biological characteristics of the three varieties.
