摘要
目的观察固本活血壮骨颗粒对糖尿病大鼠模型成骨细胞Wnt/β-catenin信号传导通路的影响。方法 6月龄SD大鼠随机分为正常对照组、模型对照组、阳性药物对照组、固本活血壮骨颗粒大、中、小剂量组。除正常对照组外,其余各组均采用左下腹腔内注射新鲜配制1%链脲佐菌素STZ(柠檬酸缓冲液,p H值4.4)65 mg/kg体重造模。各组予相应药物及生理盐水灌胃12周。结果固本活血壮骨颗粒可提高糖尿病模型成骨细胞Wnt1、LRP-5以及β-catenin蛋白的表达。结论固本活血壮骨颗粒可以通过促进成骨细胞Wnt1、LRP-5以及β-catenin蛋白的表达,调节Wnt/β-catenin信号传导通路的失调状态,促进骨形成,减少骨吸收,提高骨质量,从而达到防治糖尿病性骨质疏松症的效果。
Objective: To observe the effect of Constitution-consolidating, Blood-activating and Bone-strengthening Granules on Wnt/beta -catenin signaling pathway of osteoblast in diabetic rat model. Methods: SD rats of six months were randomly divided into normal control group, model control group, positive drug group and Granules groups of high, middle and low doses. Except the control group, the models of 65 mg/kg weight in other groups were created by injecting freshly prepared 1% streptozotocin (STZ citrate buffer, pH4.4 ) in the left abdominal cavity. The corresponding drugs and saline were given with intragastric administration for 12 weeks to the models of each group. Results: The expression of Wntl, LRP-5 andβ-catenin proteins of osteoblast significantly improved in the diabetic models by the action of Consti- tution- consolidating, Blood - activating and Bone - strengthening Granules. Conclusion: Constitution - consolidating, Blood-activating and Bone-strengthening Granules can adjust the disorder of Wnt/β-catenin signaling pathway for it can promote the expression of Wntl, LRP-5 and β-catenin protein of osteoblast, so that the diabetic osteoporosis can be pre-vented and cured because of the formation of bone, reduce of bone absorption and improvement of bone quality.
