摘要
【目的】测定树莓不同部位中鞣花酸的含量,为更合理的开发利用树莓资源提供参考依据。【方法】采用高效液相色谱法(high performance liquid chromatography,HPLC)测定树莓鲜叶、干叶和果实中鞣花酸的含量。色谱条件:色谱柱为Phenomenex Luna C18柱(250 mm×4.6 mm,5μm),以A相(0.5%磷酸水溶液)和B相(乙腈:水:冰醋酸=49.5:50:0.5)为流动相梯度洗脱,洗脱程序为:0-26 min,20%-60%B;26-30 min,60%-20%B;30-40 min,20%B。柱温30℃,检测波长254 nm,流速0.8 ml/min,进样体积20μl。【结果】鞣花酸在浓度为0.032-0.258 mg/ml范围内线性关系良好,Y=8×107X+146087(R2=0.9998)。树莓鲜叶、干叶、果实中鞣花酸的含量分别为57、76、849 mg/kg。【结论】树莓不同部位中鞣花酸的含量有较大差异,其中以果实中含量最为丰富为树叶中含量的近10倍,本研究为树莓不同部位的合理开发利用提供了实验依据。
【Objective】To determine the content of ellagic acid in different parts of raspberry and provide a reference for rational exploitation and utilization of raspberry resource.【Method】The content of ellagic acid was determined by HPLC in raspberry fresh leaves,dried leaves and fruits. The chromatogram column was Phenomenex Luna C18(250 mm×4.6 mm, 5 μm). A phase(0.5% phosphoric acid solution) and B phase(acetonitrile: water: acetic acid =49.5:50:0.5) were mobile phases for gradient elution. The gradient elution program was as follows:0-26 min, 20%-60% B; 26-30 min, 60%-20% B; 30-40 min, 20% B. The column temperature was set at 30℃.The detection was carried out at the wavelength of 254 nm. The flow rate was kept at 0.8 ml/min and the sample volume was 20 μl.【Result】The standard curve of ellagic acid was linear in the range of 0.032-0.258 mg/ml,Y=8×107X+146 087(R2=0.9998). The contents of ellagic acid in raspberry fresh leaves, dried leaves and fruits were 57, 76 and 849 mg/kg respectively.【Conclusion】The contents of ellagic acid are greatly different in different parts of raspberry. The content of ellagic acid in fruits is 10 times more than that in leaves.This study provides an experimental basis for rational exploitation and utilization of raspberry in different parts.
