冬连胶囊对STZ诱导糖尿病肾病大鼠p38MAPK、CREB1及TGFβ1蛋白表达的影响 被引量:7

摘要 目的:探讨冬连胶囊对链脲佐菌素(STZ)诱导的糖尿病肾病大鼠肾脏保护作用及TGF-β1/p38MAPK信号通路影响。方法:雄性SD大鼠60只,随机分为正常组10只,模型组50只。模型组大鼠腹腔注射链脲佐菌素(STZ)(45 mg/kg),造模成功的大鼠按体质量随机分为模型组、科素亚组、冬连胶囊低、中、高剂量组。分别于灌胃后第1、3、8周收集24 h尿液检测24 h尿蛋白定量。灌胃第8周,处死大鼠取肾组织,western-blotting法观察转化生长因子β1(TGF-β1)、p38MAPK、CREB1蛋白表达。结果:与模型组比较,各治疗组肾脏组织中TGF-β1、p-p38MAPK、p-CREB1,蛋白表达明显减少(P<0.01);冬连胶囊各治疗组可不同程度降低模型大鼠的24 h尿蛋白量,减轻模型大鼠的肾比重,对糖尿病肾病模型大鼠具有肾脏保护作用。结论:冬连胶囊可能通过调控糖尿病肾病肾组织TGF-β1/p38MAPK/CREB1信号表达,减轻肾脏细胞纤维化、减缓肾小球细胞硬化,进而降低24h尿蛋白定量,减轻肾脏损害,对糖尿病肾病起到一定的保护及延缓病程进展的作用。 Objective: To discuss the protective effect of Donglian capsules on SD rats induced by STZ and its influence on TGF-β1 / p38 MAPK signal path. Methods: Sixty SD male rats male were divided into two groups:a normal group with 10 rats and a model group with 50 rats. The rats in the model group were injected by abdominal Streptozotocin( STZ)( 45 mg / kg). After successful models,50 model rats were randomized into five groups: a model group,a Cozaar group,a Donglian low-dose group,a Donglian middle-dose group and a Donglian high-dose group. Collect 24 h urines and test 24 h urinary protein on 1,3,8 weeks after lavage. On the 8th week of lavage,kill the rats and get out kidney tissues to observe the protein expressions of TGF-β1,p38 MAPK and CREB1 by western-blotting method. Results: Compared with the model group,TGF-β1,p38 MAPK and CREB1 protein expressions of the treatment group in the kidney tissues decreased significantly( P〈 0. 01); Donglian capsules treatment groups could reduce 24 h urinary protein quantity of the STZ model rats to different degrees,and it could reduce the kidney proportion of model rats and protect kidney of diabetic nephropathy model rats. Conclusion: Donglian capsules can regulate TGF-β1 / p38 MAPK / CREB1 signal representation in kidney tissues of diabetic nephropathy,relieve renal fibrosis cells,reduce glomerular sclerosis of cells and thus reduce the 24 h urinary protein quantity,then relieve kidney damage,and it can play a protective role and delay the progression of diabetic nephropathy.
