烷基多苷在猪皮脱脂中的应用

摘要 以新鲜猪皮为原料,采用新型表面活性剂——烷基多苷对猪皮进行3步脱脂处理,运用索氏抽提法测定猪皮脱脂前后的脂肪含量,结合组织学方法分析烷基多苷对猪皮的脱脂效果,并对烷基多苷及脱脂后猪皮进行了细胞毒性检测。综合猪皮的脱脂率及组织学观察结果,得到本试验条件下猪皮脱脂的最佳工艺条件为:烷基多苷用量2%,反应温度37℃;结合MTT比色法(一种检测细胞存活和生长的方法)、高效液相色谱法对烷基多苷及脱脂处理后的猪皮进行细胞毒性检测,结果发现:烷基多苷细胞毒性的最高浓度检测限为10-6g/m L;最佳工艺条件脱脂后制得的纯化猪真皮的细胞毒性为I级。综上结果,烷基多苷可以作为一种新型脱脂材料应用在猪皮的脱脂过程中。 The new type surfactant Alkyl Polyglycoside pigskin was degreased three times in total. The content of fat in (APG) was used in degreasing process of the fresh pigskin. The pigskin was determined with the Soxhlet extraction method before and after the degreasing. The degreasing effect of APG was analysized with histological method through observing the residue of fat in pigskin. The cytotoxicity of APG and purified pigskin was tested. The results of degreasing rate and histological observation in- dicates the optimum dosage of APG is 2% of pigskin weight, reaction temperature is 37℃ in degreasing process. The testing results of MTT ( 3 - (4,5 - dimethyl - 2 - thiazolyl) - 2,5 - diphenyl - 2 - H - tetrazolium bromide) methods and high performance liq- uid chromatography (HPLC) methods show the highest concentration of cytotoxicity detection limit about APG is 10-6g/mL and the cytotoxicity of purified pigskin after optimum degreased technological condition can reach grade I. The APG can be applied to the degreasing process of pigskin as a new type degreasing material.
