摘要
目的:观察杨梅树皮氯仿萃取物对C 57BL/6J小鼠Lewis肺癌的抑制作用及对肿瘤坏死因子(TNF-α)、基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制剂-1(TIMP-1)表达的影响。方法:建立C 57BL/6J小鼠Lewis肺癌模型,分别设杨梅树皮氯仿萃取物高剂量组(13.2g生药/kg)、中剂量组(6.6 g生药/kg)、低剂量组(3.3 g生药/kg)、环磷酰胺(CTX)阳性对照组(30 mg/kg×3)及模型对照组(水30 m L/kg),给药12天后处死动物,称瘤重,计算抑瘤率(%);ELISA法测定血清TNF-α、MMP-9、TIMP-1含量。结果:杨梅树皮氯仿萃取物对C57BL/6J小鼠Lewis肺癌有一定的抑制作用,抑瘤率在16.3%~41.6%,其中高、中剂量组的瘤重与模型组比较有显著性差异(P〈0.05~0.01);同时能明显增高血清TNF-α、TIMP-1表达,降低MMP-9表达。结论:杨梅树皮氯仿萃取物对C 57BL/6J小鼠Lewis肺癌有较好的抑制作用,且能上调TNF-α、TIMP-1表达,下调MMP-9表达。
Objective: To observe the effect of chloroform extracts from Myrica rubra bark on Lewis lung cancer of mice and impact on TNF- α,MMP- 9,TIMP- 1 expressions. Methods: Lewis lung cancer models of C57 BL /6J mice were established. Mice were divided into five groups: high( 13. 2 g / kg),middle( 6. 6 g / kg)and low( 3. 3 g / kg) dose of chloroform extracts from bark of Myrica rubra,CTX group( 30 g / kg × 3) and model group( H2 O 30 m L / kg). Mice were sacrificed after 12 days. According to the final tumor weight,the inhibition rate was calculated. The expressions of TNF- α,MMP- 9 and TIMP- 1 were inspected by ELISA method.Results: Chloroform extracts from bark of Myrica rubra had certain inhibitory effect on Lewis lung cancer in mice and the inhibition rate was between 16. 3% to 41. 6%. The tumor weight of high and middle dose groups were smaller than that of model group( P 0. 05 ~ 0. 01). Chloroform extracts can also significantly increase the expressions of TNF- α and TIMP- 1,decrease the expression of MMP- 9. Conclusion: Chloroform extracts from bark of Myrica rubra had good inhibitory effect on Lewis lung cancer in C57 BL /6J mice,and can also increase the expression of TNF- α and TIMP- 1,lower the expression of MMP- 9.
