摘要
为了研究INHA基因在马关无角山羊卵泡发育中的作用,设计并化学合成了3条针对INHA基因的si RNA,脂质体法转染至马关无角山羊的卵巢颗粒细胞,Real-time PCR技术和WesternBlot技术对其干扰效率进行了筛选。结果表明:si RNA-2对INHA基因m RNA表达抑制率达到了94.3%,对蛋白表达的抑制率达到了93.4%,是INHA基因沉默的最佳干扰质粒,可为今后研究INHA基因的功能提供基础。
In order to study the role of INHA gene in the follicular development of Maguan polled goat,three si RNA against INHA gene were designed and synthesized,then these si RNA were transfected into the ovarian granular cells of goats via lipofectamine,and the interference efficiency were screened by Real- time PCR and Western- Blot. The results showed that the inhibition rate of si RNA- 2 for INHA gene m RNA expression was 94.3%, and that for protein expression was 93.4%. So si RNA- 2 is the best interference plasmid of INHA gene silencing, and which can provide the basis for the study of INHA gene function in the future.
