结肠癌转移相关基因1和肝细胞生长因子受体蛋白表达与宫颈癌盆腔淋巴结转移相关性分析 被引量:8

摘要 目的探讨结肠癌转移相关基因1(MACC1)和肝细胞生长因子受体蛋白(c-Met)表达与宫颈癌盆腔淋巴结转移相关性。方法选取宫颈癌患者113例,宫颈上皮内瘤变Ⅱ~Ⅲ患者41例,正常宫颈组织患者47例,利用免疫组化SP法检测MACC1和c-Met在组织中表达,利用二分类Logistic回归分析对宫颈癌盆腔转移相关因素进行分析。结果 MACC1和c-Met在宫颈癌组织中相对表达量分别为(7.64±1.37)和(6.73±1.22),均高于宫颈上皮内瘤变Ⅱ~Ⅲ组织和正常宫颈组织,且宫颈上皮内瘤变Ⅱ~Ⅲ组织高于正常宫颈组织,差异均有统计学意义(P〈0.05);Spearman相关分析显示,MACC1在宫颈癌组织中表达与c-Met呈正相关(r=0.467,P〈0.05);宫颈癌发生盆腔淋巴结转移与脉管浸润、肌层浸润深度、MACC1阳性表达、cMet阳性表达有关(P〈0.05),多因素Logistic回归分析显示,肌层浸润深度、MACC1阳性表达和c-Met阳性表达是宫颈癌发生盆腔淋巴结转移的独立危险因素(P〈0.05)。结论 MACC1和c-Met在宫颈癌组织中呈高表达,可能共同参与了宫颈癌发生及进展,是宫颈癌发生盆腔淋巴结转移的独立危险因素。 Objective: To explore the correlation between metastasis-associated in colon cancer-1 (MACC1), hepatocyte growth fac- tor receptor protein (c-Met) and cervical lymph node metastasis of cervical cancer. Methods: A total of 113 cervical cancer patients, 41 cervical intraepithelial neoplasia (CIN) Ⅱ-Ⅲ patients, and 47 cases with normal cervical tissue were selected. The expressions of MACC1 and c-Met were detected by immunohistochemical SP method. The related factors of cervical cancer pelvic metastasis were analyzed by binary Logistic regression analysis. Results: The relative expression levels of MACC1 and c-Met in cervical cancer were (7. 64±1.37) and (6.73±1.22), respectively, which were higher than those in CIN Ⅱ-Ⅲ tissue and normal cervical tissue, the relative expression levels of MACC1 and c-Met in CIN Ⅱ-Ⅲ tissue were higher than those in normal cervical tissue, there were statistically significant differences (P〈 0. 05) . Spearman correlation analysis showed that the expression of MACC1 was positively correlated with the expression of c-Met in cervi- cal cancer (r = O. 467, P〈O. 05) . Cervical cancer pelvic lymph node metastasis was correlated with vascular invasion, depth of myometrial invasion, MACC1 positive expression, and c-Met positive expression (P〈0. 05) . Multivariate Logistic regression analysis showed that the depth of myometrial invasion, MACC1 expression, and c-Met expression were independent risk factors of cervical cancer pelvic lymph node metastasis (P〈0.05) . Conclusion: The expressions of MACC1 and c-Met are high in cervical cancer, they might be involved in the oc- currence and progression of cervical cancer, which are independent risk factors of cervical cancer pelvic lymph node metastasis.
