全反式维甲酸减少舌癌小鼠髓样来源抑制细胞 被引量:3

摘要 目的:观察全反式维甲酸(ATRA)治疗后对机体肿瘤免疫应答内环境的改变的影响。方法:选取C57BL/6小鼠,分为3组:空白对照组10只;ATRA治疗组10只,4-硝基喹啉-1-氧化物(4NQO)水饮用16周后,腹腔给药ATRA,每周1次,连续12周;PBS组10只,4NQO水饮用16周后,腹腔注射PBS,每周1次,连续12周。第28周取全舌标本,肉眼观察大体损害,流式细胞学分析外周血中髓样来源抑制细胞(MDSC)及成熟树突状细胞(DC)的变化。结果 :整个实验期间,所有小鼠均存活。ATRA治疗显著减少了4NQO诱导小鼠舌黏膜病变发生的个数(P<0.05)。使4NQO诱导小鼠外周血中MDSC比例降低(P=0.048),成熟DC细胞比例升高,但未达到显著性意义(P=0.595)。结论 :ATRA在一定程度上可以减少舌癌小鼠体内MDSC的比例,可能增加成熟DC的比例,有助于提高机体的抗肿瘤免疫应答。 Objective To observe MDSC changes after ATRA treatment in 4NQO-induced tongue carcinoma mouse model. Methods Thirty C57 BL / 6 mice were divided into three groups: blank control group,without any treatment; ATRA treatment group, after 4NQO water treatment for 16 weeks, inject ATRA peritoneally for another 12 weeks; PBS treatment, after 4NQO water treatment for 16 weeks, inject PBS peritoneally for 12 week. Peripheral blood was collected at week 28. MDSC and mature DC expression was analyzed by flow cytometry.Results All animals survived during experimental period. After ATRA treatment, the number of lesions occurred on tongue mucosa was markedly lower(P 〈0.05). MDSC expression in the peripheral blood of ATRA treatment group was decreased than PBS treatment group(P = 0.048), and mature DC expression in ATRA treatment group was higher than PBS treatment group(P = 0.595), although such changes did not reach significance, they showed a trend. Conclusion ATRA treatment could decrease MDSC expression and increase mature DC expression in tongue tumor-bearing mice at a certain degree, which may help elevating host anticancer immune responses.
