摘要
目的检测支气管哮喘患者中外周单个核细胞Nod样受体热蛋白结构域相关蛋白3(NLRP3)mRNA和血清白细胞介素18(IL-18)、白细胞介素1β(IL-1β)的表达,探讨其在哮喘发病中的机制和临床意义。方法采用实时荧光定量聚合酶链反应(PCR)法检测100例支气管哮喘患者治疗前后和对照组外周血单个核细胞核内NLRP3mRNA的表达量,通过酶联免疫吸附测定(ELISA)法检测哮喘患者治疗前后和对照组血清IL-1β、IL-18水平,比较组间的差异,并进行相关性分析。结果急性发作期哮喘患者的NLRP3mRNA的表达及IL-18、IL-1β水平均明显高于慢性持续期哮喘患者(P<0.05)。急性发作期和慢性持续期哮喘患者的NLRP3mRNA的表达及IL-18、IL-1β水平均明显高于健康对照组(P<0.05)。急性发作期和慢性持续期哮喘患者经治疗后的NLRP3mRNA的表达量及IL-18、IL-1β水平较各自治疗前水平明显降低,且均高于健康对照组水平,差异具有统计学意义(P<0.05)。结论 NLRP3、IL-18、IL-1β均参与了支气管哮喘的气道炎症反应,且与哮喘患者的病情严度程度密切相关。
Objective To determine the expression of NLRP3 mRNA in peripheral blood monocytes( PBMCs) and IL- 1β,IL- 18 in serum,and to explore its mechanism and clinical significance in pathogenesis of bronchial asthma. Methods Quantitative real- time PCR was used to determine the expression level of NLRP3 mRNA in 100 patients with bronchial asthma before and after treatment and 50 healthy persons. Enzyme linked immunosorbent assay( ELISA) had been applied to determine the serum levels of IL- 1β and IL- 18 before and after treatment in both groups. The difference between these 2 groups was compared and the correlation analysis had been performed. Results The expression level of NLRP3 mRNA and serum levels of IL- 1β and IL- 18 in patients with bronchial asthma were higher than those of healthy controls( P〈0. 05).The expression level of NLRP3 mRNA and serum levels of IL- 1β and IL- 18 in patients with bronchial asthma in acute attack group and chronic sustained group after treatment were decreased than those before treatment,but still higher than those of normal controls( P〈0. 05). Conclusion NLRP3 and IL- 18,IL- 1β are participated in the inflammatory response in airway of patients with bronchial asthma and closely related to the severity of asthma in these patients.
