摘要
为建立规模化培养猪瘟活疫苗的生产工艺,本研究利用工作体积为10L的激流灌注式生物反应器培养猪睾丸细胞(swine testis,ST),接种猪瘟病毒(classical swine fever virus,CSFV)进行培养研究。研究结果显示,培养4 d细胞数可增长5~7倍,接种病毒后15 d可收获毒液6次,收获量至少与200个10 L转瓶单次收获量相当,产品各项检测均合格。本工艺极大地缩短了培养时间,为规模化培养工艺的推广与使用提供了技术参数。
In order to establish large-scale cultivation of swine fever vaccine production process, this research applied 10 L volume of suspension and perfusion bioreactor to culture swine testicular cells, vaccinate classical swine fever virus. Results showed that the number of cells increased 5 to 7 times within 4 days, classical swine fever virus could be harvested 6 times within 15 days, and the classical swine fever vaccine could satisfy all the biological qualifications and requirements. This technology could greatly shorten the training time and it will be the basis to such scale vaccine antigen production.
