姜黄素对于大鼠创伤性脑损伤后Bcl-2、Caspase-3蛋白表达的影响 被引量:1

摘要 目的探讨姜黄素对于创伤性脑损伤大鼠脑组织Bcl-2、Caspase-3蛋白表达的影响。方法本次研究的大鼠采用的是Wistar大鼠,选取80只并且随机分成4组,即正常对照组、创伤模型组、高剂量组(300mg/kg/d)和低剂量组(100mg/kg/d),在治疗的1天、3天、5天、7天时断髓处死各组大鼠,提取大鼠脑组织。观测指标应用免疫组织化学染色法(SP法)以及RT-PCR技术进行检测。结果大鼠脑组织受到创伤性损伤后在几天内Bcl-2、Caspase-3的蛋白表达均较正常对照组增高;姜黄素组与创伤模型组对比,其中姜黄素组Bcl-2的蛋白表达量增加,Caspase-3的蛋白表达量较少;高剂量姜黄素组的Bcl-2的蛋白表达量增加效果和Caspase-3的蛋白表达量减少效果要优于低剂量姜黄素组。结论姜黄素对于减轻大鼠的创伤性脑损伤具有一定的效果,其主要作用是促进Bcl-2的蛋白表达以及抑制Caspase-3的蛋白表达,从而达到有效减少神经细胞凋亡的效果。高剂量组效果优于低剂量组。 Objective To investigate the effect of curcumin on the expression of Bcl-2 and Caspase-3 protein in brain tissue of rats with traumatic brain injury. Methods Choose adult Wistar rats only 80,and randomly divided into four groups,namely the normal control group,wounds model group,high dose group curcumin( 300 mg / kg / d),low dose group curcumin( 100 mg / kg / d). After the therapy 1 d,3 d,5 d,7 d broken pulp put to death the rats,extraction rats organization. The observation indexes were detected by immunohistochemical staining and RT-PCR technique. Results 1. The protein expression of Bcl-2 and Caspase-3 in rat brain after traumatic injury was higher than that in normal control group within a few days after traumatic injury. 2. Comparison of curcumin group and trauma model group,the protein expression of Bcl-2 was increased and the protein expression of Caspase-3 was less in the curcumin group. 3. The protein expression increased of Bcl-2 and the protein expression to reduce of Caspase-3 in high dose curcumin group was higher than that in low dose curcumin group. Conclusion Curcumin has a certain effect on reducing traumatic brain injury in rats,the main effect is to promote the expression of Bcl-2 protein and inhibit the expression of Caspase-3 protein,so as to effectively reduce the effect of nerve cell apoptosis. The effect of high dose group was better than that of low dose group.
