摘要
在纤维素刚果红培养基中筛选具有透明圈的方法,从肥沃的土壤中筛选获得一株溶解圈直径与菌落直径比值(D/d)可达4.75的细菌,命名为DBJ。该菌最适产纤维素酶的培养时间是60 h,培养温度是35℃,起始p H 7.0。通过形态学、生理生化试验和16S r DNA序列比对分析对DBJ进行了鉴定。结果表明,革兰氏阳性菌DBJ为芬氏纤维微菌(Cellulosimicrobium funkei),将其命名为Cellulosimicrobium funkei DBJ,DBJ的16S r DNA序列的Gen Bank登录号为JX392404。
Based on the ratio of lysed zone diameter and colony diameter on the cellulose-congo red medium,a strain was isolated,its ratio was 4. 75,and designated it as DBJ. The optimum time,temperature and p H for DBJ decompose cellulose were 60 h,35 ℃ and 7. 0.Based on the morphological,physiological and phylogenetic analysis of 16 S r DNA,DBJ was identified. The result showed that DBJ was gram-positive bacterium. And DBJ was identified as Cellulosimicrobium funkei,and named Cellulosimicrobium funkei DBJ. The sequences of16 S r DNA of DBJ had been submitted to Gen Bank and the accession number was JX392404.
