细辛配伍附子含药血清对大鼠心室肌细胞L型钙通道的影响 被引量:6

Effect of Serum Containing Herba Asari and Radix Aconiti Lateralis Preparata on L-type Calcium Channel of Rat Ventricular Myocytes

摘要 【目的】观察细辛和细辛配伍附子含药血清对大鼠心室肌细胞L型钙通道的影响,探讨其抗心律失常的作用机制。【方法】连续给药7 d后制备大鼠含药血清,采用急性酶解法获得单个大鼠心室肌细胞,应用全细胞膜片钳技术记录心室肌细胞L型钙通道电流。【结果】细辛和细辛配伍附子含药血清均能增加大鼠心室肌L型钙通道电流,在0 m V钳制电压下,细辛含药血清使电流增加约(28.36±0.37)%,细辛配伍附子含药血清使电流增加(75.07±0.28)%。细辛含药血清使激活曲线、失活曲线左移,对恢复曲线无明显作用;细辛配伍附子含药血清使激活曲线、失活曲线左移,使失活后恢复时间延长。【结论】细辛配伍附子较单味细辛含药血清对心肌细胞L型钙通道的激活作用更强,表明药物配伍可增加疗效。 Objective To observe the effect of the serum containing Herbal Asari(Xixin)and Radix Aconiti Lateralis Preparata(Fuzi)on L-type calcium channel currents(ICa-L)in isolated rat ventricular myocytes,so as to investigate its antiarrhythmia mechanisms. Methods The rat serum containing Xixin and Fuzi was prepared after7-day continuous medication. Single rat ventricular myocyte was isolated by rapid enzymatic dissociation method.The whole-cell patch clamp technique was used to record the ICa-L. Results The serum containing Xixin and the serum containing Xixin and Fuzi both increased ICa-L. At 0 m V voltage,the serum containing Xixin increased the ICa-Lby(28.36±0.37)%,and the serum containing Xixin and Fuzi increased ICa-Lby(75.07±0.28)%. The serum containing Xixin induced left shift of the activation curve and inactivation curve,but had no effect on recovery time curve. The serum containing Xixin and Fuzi also induced the left shift of the activation curve and inactivation curve, and prolonged the recovery time after inactivation. Conclusion The compatibility of Xixin and Fuzi augments the ICa-Lactivation much more than Xixin alone, indicating that drug compatibility may increase the therapeutic efficacy.
