摘要
【目的】观察益气活血化痰汤对溃疡性结肠炎(UC)模型大鼠结肠黏膜血管内皮细胞生长因子(VEGF)和细胞间黏附分子-1(ICAM-1)基因表达的作用。【方法】采用三硝基苯磺酸(TNBS)法复制UC大鼠模型,将60只大鼠分为正常组,模型组,中药高、中、低剂量组(剂量分别为52、26、13 g·kg^(-1)·d^(-1))和美沙拉嗪组(剂量为0.5 g·kg^(-1)·d^(-1)),分别检测各组大鼠体质量、结肠黏膜病理变化,以及肠黏膜VEGF、ICAM-1基因表达水平。【结果】模型组体质量显著降低,结肠黏膜出现明显病理改变,VEGF和ICAM-1基因相对表达量均显著高于正常组(P<0.01)。中药各剂量组均可显著升高大鼠体质量,改善结肠黏膜病理变化,显著降低VEGF、ICAM-1表达水平(P<0.01),其中中药高剂量组作用更佳。【结论】益气活血化痰汤对VEGF和ICAM-1基因表达有下调作用,这可能是其治疗UC作用的机理之一。
Objective To investigate the effect of Yiqi Huoxue Huatan Decoction(YHHD)on gene expression of vascular endothelial growth factor(VEGF),intercellular adhesion molecule 1(ICAM-1)in the colonic mucosa of ulcerative colitis rats. Methods Sixty rats were divided into six groups, namely normal control group, model control group,low-,middle- and high-dose Y HHD group(gastric gavage of YHHD in the dosage of 13,26,52g·kg-1·d-1respectively), and Mesalazine(0.5 g·kg-1·d-1) group. T he rat model of ulcerative colitis was established by enema with 2,4,6-trinitrobenzene sulfonic acid(TNBS)mixture(2.5% TNBS and 50% ethanol).The changes in rat body weight were monitored. After treatment, pathological features of the colonic mucosa were observed under light microscope, and VEGF and ICAM-1 gene expression levels in the colonic mucosa were measured by reverse transcription polymerase chai n reaction(RT-PCR). Results Body weight was lighter,and relative gene expression levels of VEGF and ICAM-1 in the model group were significantly higher than those in the normal control group,and pathological features of the colonic mucosa was obvious,the differences being significant between the two groups(P〈0.01). Compared with the model group,body weight was increased,pathological features of the colonic mucosa were relieved, and relative gene expression levels of VEGF and ICAM-1 were significantly decreased in YHHD groups(P〈0.01),and high-dose YHHD showed the strongest effect. Conclusion YHHD can down-regulate the gene expression of VEGF and ICAM-1,which may be one of its therapeutic mechanisms for ulcerative colitis.
