摘要
目的:探讨外源性前列腺素E_2(PGE_2)通过影响整合素αv、β3对胚胎黏附率的改变及可能的机制。方法:使用实时定量聚合酶链反应(real-time PCR)、蛋白质印迹(Western blotting)检测外源性给予PGE_2后,RL95-2细胞的整合素αv、β3 m RNA及蛋白表达水平的变化。利用Be Wo滋养层细胞和RL95-2上皮细胞株建立子宫内膜-滋养细胞相互作用的体外模型,观察Be Wo细胞球在经过不同浓度PGE_2预处理的RL95-2上皮细胞单层的黏附效率。结果:200 nmol/L PGE_2预处理RL95-2细胞单层后促使整合素αv的m RNA及蛋白表达量比0 nmol/L组增加(均P<0.05);100 nmol/L PGE_2预处理RL95-2细胞单层后促使整合素β3的m RNA及蛋白表达量比0 nmol/L组增加(均P<0.05)。200 nmol/L PGE_2预处理后Be Wo细胞球黏附到RL95-2细胞单层的效率明显提高。结论:适当浓度的PGE_2可上调整合素αv、β3的表达,促进胚胎黏附效率。提供了在植入期间促进整合素αv、β3表达的进一步证据,这将有助于阐明通过改善子宫内膜容受性促进胚胎黏附的分子机制。
Objective:To explore the effect of exogenous prostaglandin E_2 on the embryonic adhesion rate by integrin αv and β3, as well as the potential mechanism. Methods:The expressions of integrin αv and β3m RNA and protein in the cultured RL95-2 cells treated with exogenous PGE_2 were detected by real-time PCR and Western blotting. Be Wo trophoblast cells and RL95-2 endometrial epithelial cells were used to establish the in vitro model. The RL95-2 epithelial cell monolayer was pretreated by different concentrations of PGE_2. The rate of the adhesion between Be Wo cell and RL95-2 epithelial cell monolayer was compared. Results: PGE_2(200nmol/L) significantly increased the expressions of integrin αv m RNA and protein(P〈0.05), while PGE_2(100nmol/L) also significantly increased the expressions of integrin β3 m RNA and protein in RL95-2 cells(P〈0.05). The rate of the Be Wo sphere cell adhered to the RL95-2 cell monolayer was significantly increased by the pretreatment with 200 nmol/L PGE_2. Conclusions: PGE_2 in appropriate concentration may increase the expressions of integrin αv and β3, and improve the embryonic adhesion. This study provides further evidence to promote the embryonic adhesion during implantation by increasing the expression of integrin αv and β3 and improving the endometrial receptivity.
