摘要
目的:观察电针对新生大鼠缺氧缺血性脑病模型(HIE)mdm2表达的干预机制。方法:7 d龄SD大鼠随机分为假手术组、模型组、假电针组、电针组、拮抗剂组、电针+拮抗剂组,各组又分为1 d、3 d、7 d、21 d四时段,结扎双侧颈总动脉,建造HIE模型。透射电镜观察脑组织超微结构;RT-PCR检测mdm2 m RNA转录水平;Western blot检测mdm2蛋白表达。结果:电镜显示21 d电针组神经元超微结构损伤明显减轻,胞膜恢复完整,染色质尚均匀,偶见肿胀的线粒体。电针组mdm2蛋白表达均下调,与模型组相比,有统计学意义(P<0.05)。结论:电针可明显减轻神经元超微结构退行性变化,其作用机制可能与mdm2表达下调有关,对HIE大鼠发挥神经保护作用。
Objective: To explore the effect of electroacupuncture on the expression of mdm2 following hypoxic-ischemic encephalopathy( HIE).Method: The hypoxic-ischemic models were made in new-bore SD rats aged 7 days by colligating bilateral common carotid artery. All rats were randomly divided into sham operation,model,sham acupuncture,electroacupuncture,antagonist and electroacupuncture with antagonist groups which were subdivided into 1 d,3 d,7 d,21 d time-phases. Ultra-structure of neurons was observed by using TEM and real-time PCR was used to determine the level of mdm2 m RNA and the expression of mdm2 was tested with Western Blot.Results: TEM showed that on 21 d,the damnification of neurons had evidently comeback with electroacupuncture treatment. The down-regulation of mdm2 was observed in electroacupuncture group,which was significantly lower than those in model groups( P〈0. 05). Conclusion: Electroacupuncture could obviously lighten the damage of ultrastructure of neurons to protect the neurons from HIE by inhibiting the expression of mdm2.
