小叶丁香总皂苷对衰老模型小鼠的抗氧化作用研究 被引量:3

摘要 目的:考察小叶丁香总皂苷对D-半乳糖致衰老模型小鼠抗氧化作用。方法:采用小鼠颈背部皮下注射D-半乳糖溶液200 mg·kg-1·d-1建立衰老模型;造模成功后,将小鼠随机分为模型对照组、小叶丁香总皂苷低剂量组(60 mg·kg-1)、小叶丁香总皂苷高剂量组(120 mg·kg-1)和阳性药对照组(维生素E,50 mg·kg-1),连续给药8周后检测血及脑组织中的超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)和总抗氧化能力(T-AOC)等指标。结果:与模型对照组比较,小叶丁香总皂苷低剂量组能明显升高血清中的SOD、GSH-Px和CAT活性以及脑组织中的T-AOC活性;小叶丁香高剂量组能显著降低小鼠血清及脑组织中的MDA水平,升高小鼠血清中的SOD、GSH-Px、CAT和T-AOC活性以及脑组织中的SOD、GSH-Px和T-AOC活性。结论:小叶丁香总皂苷能抑制机体内脂质过氧化物的生成,增强抗氧化酶的活性,从而提高衰老小鼠的抗氧化能力,延缓衰老。 Objective: To investigate the effect of total saponins from Syringa pubescens on the antioxidantive property in D-galactose-induced aging mice. Methods: Each ICR mouse daily received cervix subcutaneous injections of 200 mg·kg-1D-galactose for 8 weeks to establish aging model. Successful model mice were treated with vitamin E and total saponins from Syringa pubescens( 60 mg · kg-1and 120 mg · kg-1) for 8 weeks. The indicators included superoxide dismutase( SOD),malondialdehyde( MDA),glutathione peroxidase( GSH-Px),hydrogen peroxidase( CAT) and total anti-oxidatant capacity( T-AOC). Results: Compared with aging control,the total saponins from Syringa pubescens( 60 mg·kg-1) significantly increased serum SOD,GSH-Px and CAT activities and brain T-AOC activities in aging mice. The total saponins at 200 mg·kg-1reduced MDA level in serum and brain tissue and increased serum of SOD,GSH-Px,CAT and T-AOC activities and brain SOD,GSH-Px and T-AOC activities in aging mice. Conclusions: The total saponins from Syringa pubescens can inhibit the generation of lipid peroxides and increase antioxidant enzyme activities so that it can significantly increase the antioxidant ability in the body and delay the aging process in mice.
