摘要
以大果黑果枸杞嫩茎为外植体,以MS为基本培养基,研究激素组合对大果黑果枸杞组培苗初代、继代以及生根培养的影响,建立大果黑果枸杞组培快繁技术体系。结果表明:适合大果黑果枸杞初代培养的培养基为MS+ZT 0.2 mg·L-1+IBA 0.01 mg·L-1,腋芽生长良好,萌芽率达88.73%,且玻璃化情况很少;ZT对组培苗增殖培养的效果优于6-BA,适合组培苗增殖培养的培养基为MS+ZT 0.15 mg·L-1+IBA 0.01 mg·L-1,组培苗生长速度快,增殖倍数可达5.83倍,基本没有玻璃化现象,且苗生长健壮;适合组培苗生根的培养基为MS+IBA 1.0 mg·L-1,生根率达100%。
The purpose of this experiment was establishing the technical system of tissue culture and rapid propagation of Lycium ruthenicum.In this study,tender stem of Lycium ruthenicum as explant,MS as basic culture medium,the effects of different factors on primary culture,subculture and rooting culture were studied. The results showed that: the suitable medium for axillary bud sprouting was MS+ZT 0.2 mg·L-1+IBA 0.01 mg·L-1,in which the axillary bud germination rate reached 88.73% and the vitrification were rarely;in the same time,the effect of ZT on the proliferation of tissue cultured plantlets was better than that of 6-BA.The suitable medium for proliferation was MS+ZT 0.15 mg·L-1+IBA 0.01 mg·L-1,in which the plantlets grew quickly,the proliferation rate was 5.83 times,no vitrification,seedling robust;the suitable medium for plantlet rooting was MS+IBA 1.0 mg·L-1,in which the rooting rate reached 100%.
