昼夜节律基因PER1对成骨相关基因表达影响的体外研究

The effects of circadian rhythm gene PER1 on the expression of osteogenesis related genes in vitro

摘要 目的:研究体外成骨细胞中,昼夜节律基因PER1对成骨相关基因表达的影响。方法:体外培养小鼠成骨细胞系MC3T3-E1,加入地塞米松干预,按时间节点0 h、2 h、4 h、8 h、12 h、16 h、20 h、24 h收集细胞,使用荧光定量PCR检测Per1(Period1)和成骨细胞相关基因碱性磷酸酶(ALP)、骨钙素(OC)和基质金属蛋白酶2(MMP-2)的表达。结果:使用地塞米松干预后,Per1基因表达在8 h达到峰值,约为初始表达量的9倍。成骨细胞相关基因ALP表达在8 h出现峰值;成骨细胞相关基因OC在12 h达到峰值,为初始表达量的20余倍;成骨细胞相关基因MMP2表达量明显降低,4 h出现低值后缓慢增加,12 h达到与原始值持平。结论:使用地塞米松干预MC3TE-E1细胞后,成骨细胞相关基因ALP、OC、MMP2的表达量会随Per1基因表达量发生相应的改变。 Objective:To explore the relationship of relative expression between per1 gene and osteogenesis related gene. Method:Culturing MC3T3-E1 cells in vitro,treating them with dexamethasone,collecting these cells at 0 h,2 h,4 h,8 h,12 h,16 h,20 h,24 h and then using quantitative real time PCR to quantify them. Result:After using dexamethasone,Per1 gene's repression reached the peak after 8 h,and was nine fold of initial value. Osteogenesis related gene ALP's repression reached the peak after 8 h. Osteogenesis related gene OC's repression reached the peak after 12 h,equaled twenty fold of initial value. Osteogenesis related gene MMP2's repression sharply declined. It reached the valley at 4h and then increased slowly to the initial value at 12 h. Conclusion:After treating MC3T3-E1 cells with dexamethasone,osteogenesis related genes' expression level,including ALP,OC and MMP2,changed with the level of PER1 gene's repression level.
